Abstract


Objective: chickpea isoflavone extraction process of total research and explore chickpea isoflavone extract (CI) for the prevention of hyperlipidemia mice . Methods: ( 1 ) to Xinjiang estate chickpeas as raw material, degreasing, ethanol extract , ethyl acetate, to obtain total isoflavones , using orthogonal design the best technology . ( 2 ) to CI, the test substance , the use of 78 male Kunming mice were randomly divided into six groups: normal control group (NC), hyperlipidemia model group (HC), CI high, medium and low dose Zhibituo treatment group and the groups of 13 . In addition to the NC group fed with distilled water daily morning , each group were fed with high fat emulsion 0.5mL / only · d. Meanwhile three dose groups were fed with in the afternoon CI640, 320,160 mg / Kg.d, Zhibituo treatment group were fed Zhibituo capsules 75 mg / Kg.d, NC and HC groups to give the corresponding plasmin for 10 days. Weighed every three days , end of the experiment serum total cholesterol (TC), triglyceride (TG) and high-density lipoprotein cholesterol (HDL-C), liver superoxide dismutase (SOD) and malondialdehyde ( MDA), and take a leaf made ??after pathological examination of liver tissue fixation . Results : Screening optimum ethanol extraction conditions were : five times the amount of 60% ethanol extracted four times, each time one hour ; ethyl acetate , and the resulting total isoflavone content of 22.15% . CI for high cholesterol was significantly lower serum TC effect ; pair had elevated HDL-C effect ; may be elevated liver SOD activity and lower MDA content . Conclusion: chickpeas mixed solvent used for the extraction of raw materials isoflavones method is feasible. CI mice on high fat diet -induced hyperlipidemia has a good preventive effect, particularly in the cholesterol-lowering effect is most obvious mechanism may be inhibition of lipid peroxidation .
