Abstract


Objectives:1. To establish the obese rat model induced by high-fat diet.2. To study and observethe differences in lipid level, serum adiponectin and vascular endothelial NF-kB ofhigh-fat induced dietary obese rats, after administration of lactobacillus acidophilus andfructo-oligosaccharide.3. To explore the effect of microeconomics on body weight,lipid level, serum adiponectin and expression of vascular endothelial NF-κB on obeserats.Methods:40male SPF SD (Sprague Dawley) rats (provided by The Experimental AnimalCenter of Dalian Medical University) which were3weeks old and50±5g in weightwere randomly divided into two groups:control group and experimental group.8rats ofcontrol group fed with fat free diet and experimental group32were fed with high-fatdiet. Both groups could access drinking water without restriction and weighed weekly.The experiment was a success as it was marked that in four weeks time the weight of25rats of experimental group was20%higher compared to control group. The successfulexperimental group rats were further divided into3groups of8rats each: group A fedwith high-fat diet, group B fed with high-fat diet and administered with lactobacillusacidophilus1ml/100g in stomach, group C fed with high-fat diet, administered withlactobacillus acidophilus1ml/100g and fructo-oligosaccharide400mg/100g in stomach.Group A and control group N fed with normal saline in the standard of1ml/100g, andall groups weighed weekly for weeks. After8weeks of experiment, the blood sampleswere taken from punctured left heart of the rats and centrifuged for analysing of lipidlevel, serum adiponectin by enzymes linked immunosorbent assay (ELISA). NF-κBexpression in vascicular endothelial of thoraco-abdominal aorta was detected byimmunochemistry after the body perfused with paraformaldehyde, then data was analyzed by SPSS17.0software.Results1.The rats of experimental group increased in weight and scarcely moved aroundcompared to the rats of control group.8weeks later,the average weight of group A was471.0±9.0g, group B450.4±4.5, group C402.2±8.38g, control group N385.2±5.0g.Theweight of group A was higher than any other (P <0.05). Among group B, C, N, theweight of group B was the highest (P<0.05).The weight of group C was the least (P>0.05).2.Lipid level: the TC of group A,B,C,and N were69.2±12.8ummol/L,52.4±3.6ummol/L,43.8±3.1ummol/L,43.3±4.8ummol/L,respectively. The TG of group A, B,Cand N were44.6±5.8ummol/L,25.0±5.5ummol/L,23.2±3.1ummol/L,16.6±4.6ummol/L. The LDL-C of group A, B,C,and N were34.8±6.6ummol/L,26.4±3.8ummol/L,20.6±3.6ummol/L,15.8±4.5ummol/L. The HDL-C of groups A, B,C andN were11.6±2.2ummol/L,13.0±3.2ummol/L,12.4±2.1ummol/L,16.6±4.1ummol/L.In group A the value of TC, TG, LDL-C`s levels were higher than those in control groupN (p<0.05). The value of TC, TG, LDL-C in group B were lower than of group A(P<0.05).The value of TC, TG, LDL-C in group C were lower than of group A(p<0.05).The value of TC, LDL-C in group C levels were lower than those of group B(p<0.05).While the TG value of group C was lower than the value of group B(P<0.05).The value of TC,TG,LDL and HDL of group C compared with control group N, thevalue of TG was higher than that of control group N (p<0.05).3.The level of serum adiponectin: the levels of serum adiponectin of group A, B, Cand N were0.60±0.08ng/ml,0.78±0.07ng/ml,0.91±0.06ng/ml,1.0±0.12ng/mlrespectively. The level of group A was the lowest (P<0.05). Amongst group B, C andN,the level of serum adiponectin of group B was the lowest (p<0.05), the level ofserum adiponectin of group C was slightly lower than of group N(p>0.05).4.The relationship between serum adiponectin and Lipid level:Adiponectin wasnegatively correlated with triglyceride,while there was no relationship between serumadiponectin and low density lipoprotein,cholesterol, total cholesterol, high-densitylipoprotein cholesterol.5.The expression of NF-κB in vascular endothelial: immunohistochemistrydisplayed that NF-κB mainly stains in vascular endothelial, a membrane of smoothmuscle cell and some cytoplasma of smooth muscle cell. The NF-κB positive expression rates and scores of group A,B, C and N were92.5%(37/40) and3.24±0.28,67.5%(27/40)and2.16±0.13,67.5%and2.16±0.13,32.5%(13/40) and0.49±0.17respectively. The NF-κB positive expression rate and score of group A was the highest(p<0.05). Amongst group B, C and N, group N was the lowest (p<0.05). Group B washigher than group C (p>0.05).Conclusion1.The prevalence of obesity is increasing, the consumption of high-fat diet resultsin overweighed SD rats that can be ideally used as models after4weeks. After8weeksof same diet lead to higher level of lipids, lower serum adiponectin and higherexpression of vascular endothelial NF-κB.2.A mix of lactobacillus acidophilus and fructo-oligosaccharide decelerates betterthe increase in weight of obese rats than if used separately.3.If lactobacillus acidophilus and fructo-oligosaccharide consumed together on anobese rat has a better effect on reducing lipid levels and decreasing the endothelialNF-κB positive expression while increasing the serum adiponectin levels than whenused separately.
