Change of the Production of INF-γ, IL-10 in Peripheral Blood Mononuclear Cells from Convulsion Rats and the Influence of Immunoglobulin on it

Abstract


Research purposes : First, to investigate whether repeated seizures cause dysfunction of the rat T cell subsets , performance mononuclear cells (PBMC) secrete interferon γ (INF-γ) and interleukin -10 (IL-10) to change ; Second imbalance of T cell subsets regulation to further clarify the role of the antiepileptic and neuroprotective the immunomodulatory mechanism to provide a theoretical basis for IVIG treatment of epilepsy , research immunoglobulin (IVIG) . Methods: repeatedly by intraperitoneal injection of pentylenetetrazol (PTZ) establish a rat model of epilepsy by mononuclear cell culture technology , the supernatant of IL-10 and INF-γ concentration was measured by ELISA . Results: 1 . Seizure group than non- seizure group culture supernatant of IL-10 and INF-γ concentration was significantly higher ( P lt; 0.001 ) , and the convulsions of IL-10 with both the level of INF-γ was positively correlated correlation coefficient 0.536, P = 0.015 , at the same time , the seizure group than non- the seizure group IL-10/TNF-γ ratio was significantly lower ( P lt; 0.001 ) . IVIG culture . Convulsions group than IVIG culture supernatant of IL-10 concentration was significantly higher ( P lt; 0.001 ) ; INF-γ concentration was significantly lower ( P lt; 0.001 ) . Conclusion: 1 , repeated seizures can lead to the dysfunction of the rat T cell subsets . Immunoglobulin may have a role in regulating T cell subsets disorders , to think that this can be one of the mechanisms for antiepileptic and neuroprotective effects .
