Abstract


Objective: To investigate the the hPOT1, p53 gene in gastric cancer clinical pathological features the silencing of hPOT1 in the gastric tissue, p53 protein and mRNA and HP infection detection, relationship and meaning, and HP may cause cancer mechanism. Methods: (1) by immunohistochemical SP method detected 53 cases of gastric carcinoma and 10 cases normal gastric mucosa silencing of hPOT1 of p53 protein expression. (2) application of in situ hybridization method detected 53 cases of gastric carcinoma and 10 normal gastric mucosa hPOT1mRNA of p53 mRNA expression. (3) Application of Warthin-Starry silver staining, detection of HP infection in 53 cases of gastric carcinoma. Results: (1) the protein expression hPOT1 53 cases of gastric carcinoma was 84.91% (45/53), the positive rate was significantly higher than 30% of normal gastric mucosa (3/10), the difference was between the two significance (P lt; 0.01); gastric carcinoma hPOT1 protein positive expression rate of the patient's age, sex, location and lymph node metastasis was no significant correlation (P gt; 0.05), but with gastric cancer histological type and differentiation The degree of progress on the degree of pathological changes significantly (P lt; 0.05), signet ring cell carcinoma and mucinous adenocarcinoma than gastric papillary and tubular adenocarcinoma hPOT1, protein expression was significantly higher, poorly differentiated gastric cancer compared with undifferentiated carcinoma of the stomach high school hPOT1 protein expression was significantly increased in advanced gastric cancer compared with early gastric cancer hPOT1 protein expression was significantly higher. (2) the positive expression of p53 protein in 53 cases of gastric carcinoma was 60.38% (32/53), the positive expression rate was significantly higher than that in normal gastric mucosa, 10% (1/10), there was significant difference between the two ( P lt; 0.05); gastric carcinoma p53 protein positive expression rate of the patient's age, gender, location, lymph node metastasis, histological type and degree of differentiation was no significant correlation (P gt; 0.05), but with gastric The progress of the pathological changes were significantly correlated (P lt; 0.05), in advanced gastric cancer group p53 protein expression was significantly higher than that of early gastric cancer. (3) in 53 cases of gastric carcinoma, the positive expression rate of hPOT1mRNA 58.49% (31/53) the normal mucosa hPOT1mRNA positive expression rate of 10% (1/10), between the two, the difference was statistically significant (P lt; 0.05); gastric carcinoma hPOT1mRNA positive expression rate of the patient's age, gender, location, lymph node metastasis, histological type and degree of differentiation was no significant correlation (P gt; 0.05), but with changes in gastric pathology progress were significantly correlated (P lt; 0.05), in advanced gastric hPOT1mRNA positive expression rate was significantly higher than that of early gastric cancer. (4) in 53 cases of gastric carcinoma p53mRNA positive expression rate of 30.20% (16/53), the positive rate was significantly lower than that in normal gastric mucosa, 70% (7/10) between the two, the difference was statistically significant (P lt; 0.05). (5) in 53 cases of gastric cancer, HP infection was positive in 28 cases (52.83%), HP infection negative in 25 cases (47.17%). (6) in 53 cases of gastric carcinoma common hPOT1 p53 protein positive expression rate was 58.4% (31/53), statistical analysis, both in gastric carcinoma were positively correlated (r = 0.416, P = 0.002); 53 cases of gastric carcinoma hPOT1 protein and hPOT1mRNA of common positive expression rate was 56.60% (30/53), statistical analysis, both in gastric carcinoma were positively correlated (r = 0.394, P = 0.004); 53 cases of gastric cancer The positive expression rate of p53 protein with p53mRNA of common organization is 7.55% (4/53), statistical analysis of both gastric expression was negatively correlated (r = -0.476, P = 0.000). (7) in 53 cases of gastric cancer, HP positive group hPOT1 infection protein positive expression rate was 96.43% (27/28), significantly higher than the HP infection negative group, 73% (18/25) (P lt; 0.05); HP infection positive group p53 protein expression was 82.12% (23/28) was significantly higher than the the HP infection negative group, 36% (9/25) (P lt; 0.01); the HP infection positive group hPOT1mRNA positive expression rate was 85.75% (24 / 28), significantly higher than the HP infection negative group, 28.00% (7/25) (P lt; 0.01). Conclusion: (1) gastric tissue the hPOT1 Protein and 1mRNA of expression levels with the increase in the degree of gastric malignant increase the expression of hPOT1 Protein and 1mRNA of gastric cancer malignancy and prognosis of gastric. (2) in gastric carcinoma abnormal expression of p53 protein levels associated with the development of gastric carcinogenesis, missing wild-type p53mRNA expression may participate in gastric carcinogenesis. (3) of mutant p53 hPOT1 in the incidence of gastric cancer, the development of a synergistic effect. (4) HP infection in gastric tissue caused hPOT1 and p53 abnormal expression may be one of the mechanisms that lead to gastric cancer.
