Abstract


Objective: To investigate the therapeutic effect of topical betaxolol eye drops on experimental retinal ischemia-reperfusion injury and its possible mechanism . Methods: the paracentesis pressure perfusion increased intraocular pressure to 110mmHg maintain 60min production in rat experimental retinal ischemia-reperfusion injury model . 56 healthy SD rats were randomly divided into normal control group (N = 8), ischemia -reperfusion 0.25% betaxolol group ( I / R BE group , 24 ) , ischemia-reperfusion saline group ( I / R NS group , 24 ) . The latter were divided into two groups according to the different time of reperfusion 1,3,7 d groups of 8 rats , each group of mice right eye ischemia-reperfusion eye . The beginning of reperfusion and every morning and evening ( 9 - 10a.m and 8 - 9p.m . ) , I / R BE group mouse right eye instillation of 0.25% betaxolol eye drops , I / R NS group rat right eye instillation of saline . 1,3,7 d, respectively , after reperfusion optical microscopy and transmission electron microscopy under observation the retina Pathomorphology and ultrastructural changes , ERG ( ERG ) recording the change of the b-wave amplitude , immunohistochemistry ( SABC method expression of nNOS ) detection of retinal tissue optical density (OD) . Results: perfusion time , the I / R the BE group of retinal morphology and ultrastructure damage than the I / R NS group was significantly reduced ; I / R BE group ERG b -wave amplitude recovery percentage recovery ( 57.24 ± 4.54,73.77 ± 9.33 , 89.23 ± 6.75 ) was significantly higher than the I / R NS group ( 32.13 ± 7.28,23.91 ± 2.77,16.51 ± 8.38 ) ( P lt ; 0.01 ) ; N group inner nuclear layer and ganglion cell layer a small amount of nNOS expression , I / R NS group of nNOS expression 1d began to increase after reperfusion , reperfusion 3d reached the peak , I / R the BE group of nNOS OD value (0.657 ± 0.013,0.684 ± 0.058,0.670 ± 0.026) was significantly lower than the I / R NS group (1.573 ± 0.353,3.740 ± 0.118,2.460 ± 0.603) (P lt; 0.01), compared with N group , no significant difference ( 0.638 ± 0.0421 ( P GT ; 0.05 ) Conclusion: topical betaxolol eye drops on experimental retinal ischemia -reperfusion injury has a protective effect may be reduced with betaxolol intracellular Ca is 2 overload , inhibit the role of free radicals .
