In this study, the antisense RNA probe in situ hybridization analysis of vasa gene expression in the eel gonads natural reversal process . The results showed that : the vasa mRNA Ⅰ , Ⅱ, Ⅲ of oocyte cytoplasm uniform distribution Ⅳ Ⅴ of oocytes , vasa mRNA concentration of migration to the cytoplasm peripheral cortex , but it is not obvious ; degradation of eggs also showed the vasa mRNA positive reaction ; Ⅲ, Ⅳ stage ovarian capsule detected with vasa positive signal cells , these cells may be differentiated spermatogonia , primordial germ cells migrate to the ovarian membrane (PGC); to these PGC in the process of sex reversal may be caused by ovarian capsule migrate to fine lobules concurrent bred sperm ; mature testis , vasa expression in spermatogonia and primary spermatocytes . Further use of the alkaline phosphatase staining method analysis albus ovary and testis revealed that: in the ovary , in addition to the oocytes , ovarian film detected with alkaline phosphatase positive signal cells ; only in mature testis alkaline phosphatase, detected in the male germ cells of the gonad sac testis film was not detected in cells with alkaline phosphatase - positive signal . The preliminary results of this study show that : eel male germ cells may originate in the primordial germ cells in the female stage ovarian capsule .
