Abstract


Background and purpose: ursolic acid (Ursolic acid, UA), triterpene acids, exist in a variety of plants in nature, with a wide range of biological activity. Modern scientific research has found that ursolic acid has hepatoprotective, antibacterial, anti-inflammatory, and lipid-lowering anti-artery atherosclerosis, immune regulation, and promote functional recovery of the hematopoietic system and the multiple functions of anti-tumor. In recent years, ursolic acid anti-tumor effect of increasing attention, the study found that ursolic acid has a variety of tumor cell lines growth inhibition, cytotoxicity and induction of apoptosis, but its mechanism is not yet clear. This study was to investigate the Ursolic acid inhibition and induction of apoptosis mechanisms in vitro on human gastric cancer cell proliferation of BGC-823. Methods: The human gastric cancer cell line BGC-823 were cultured with RPMI-1640. BGC-823 cell proliferation using the MTT assay ursolic acid inhibitory effect; using agarose gel electrophoresis DNA apoptotic fragments; BGC-823 cells by flow cytometry detection of ursolic acid cycle changes and drug effects the rate of apoptosis, and BGC-823 cell surface Fas expression change; Western blot Western blotting detection BGC-823 cells Bcl-2 expression and caspase-3 and caspase-8 activity changes. Results: 1.MTT results show that ursolic acid inhibit the proliferation of BGC-823 cells, and a dose-and time-dependent manner. Ursolic acid for 24h IC50 value 43.10μmol / L. 2 gel imaging system under observation showed no obvious DNA ladder formation 0,20,30,40 μmol / L ursolic acid for 24h, and 50μmol / L, 60 μmol / L ursolic acid effect 24 hours after the electrophoresis The band performance the characteristic ladder was found with (DNA ladder). Cell cycle detected, with the increase in the concentration of ursolic acid, BGC-823 cell subsets G 1 peak gradually increased, G 1 period of decline in the S phase arrest increased G 2 / M phase no significant change; gradual increase in the concentrations of these drugs, the percentage of apoptosis of BGC-823. Flow cytometry showed low BGC-823 cell surface expression of Fas (0.92%), ursolic acid role BGC-823 cell surface expression of Fas after 24h no significant change. 5.Western blot blotting results show that ursolic acid for 24h with increasing drug concentration, Bcl-2 protein expression decreased gradually; decreased the expression of caspase-8 zymogen 43kD 41 kD intermediate and 18kD active fragment increased expression; of Caspase-3 as the ursolic concentration increases, the decline in the expression of zymogen, increase in the expression of the 17kD and 19kD active fragment.
