Abstract


Objective To compare the cytokine-induced peripheral blood sources idiopathic thrombocytopenic purpura (ITP) in patients with dendritic cells (DC) and normal DC difference in immune molecule expression and immune function , and explore the outer periphery of the ITP patients DC in which the functional status of the blood , and speculated DC through its role in the pathogenesis of ITP . 38 cases of the method clinically diagnosed ITP patients , collected from ITP patients in vivo 15ml peripheral blood lymphocyte separation medium separation and centrifugal interface cells adherent culture 2h mononuclear cells added cytokines that rh GM- CSF ( 100ng/ml) and rhIL -4 ( 500U/ml ) and LPS (50U/ml), and expanded in vitro for six days , in the light microscope to observe the growth . After 6 days of culture , cell morphology was observed with scanning microscopy and transmission microscopy ; FITC- CD80 and PE-labeled CD86 monoclonal antibody labeled DC, DC phenotype determined by flow cytometry of CD80 , CD86 expression level ; MTT assay ITP patients with DC to stimulate allogeneic T cell proliferation ; the DC research methods such as IL-12 secretion was measured to IL-12ELISA kit . 23 cases of healthy adults as a control . Results the 5 celestial cultured DC suspension burr-like state becomes adherent cells , with prolonged incubation time , LPS was added after 6 days , such an increase in cell number , for irregular burr-like , for typical dendritic like cell morphology . Flow cytometry showed that patients with ITP mature DC express high levels of CD80 and CD86 molecules . MTT assay ITP patients DC and normal DC to stimulate allogeneic T lymphocyte proliferation , ITP patients to stimulate allogeneic lymphocytes proliferation was markedly stronger than the normal control group . DC secreted supernatant ELISA kit , training six days , idiopathic thrombocytopenic purpura patients DC secreted IL-12 levels higher than normal . Conclusion ① human peripheral blood induced by rh GM-CSF rh IL-4 LPS in vitro culture can induce a mature DC ② ITP patients with peripheral blood culture in DC than normal peripheral blood mononuclear cells of the mature state of ③ ITP patients prefer to DC differentiation DC with respect of the ability to enhance the differentiation from normal subjects with a strong antigen-presenting ability .
