Abstract

Background and Purpose: With the improvement of medical standards, changes in the human disease spectrum in patients with severe and immune suppressed patients on the rise, Candida infections of these two types of patients are more prone to nosocomial infections, the treatment of a limited range of medicines. In recent years, the development of antifungal drug made great progress, but fluconazole and itraconazole is still the most commonly used clinical drug use has been nearly 20 years since the 1990s. Other areas of foreign and domestic research has been reported in the past 20 years, the clinical Candida sub outrageous changed, increased azole drug resistance phenomenon, but the city of Wuhan to the lack of research in this area information. The purpose of this project is to get outrageous points of of Wuhan city pathogenic yeast, clinical different kinds of isolates to fluconazole, itraconazole vitro drug sensitivity. Methods: Candida separation specimens Wuhan three large general hospital inpatients (People's Hospital of Wuhan University, Zhongnan Hospital, Wuhan University, Huazhong University of Science and Technology, Tongji Hospital) from October 2003 to April 2004. Low temperature preserved, YBC chromogenic card Microbial analyzer Candida identification to the species level. The transfer species survive the strains and standard quality control strains centralized by disk diffusion method M38-A program released by the American National Clinical Laboratory Standards Commission and microdilution method M27-A2 scenario fluconazole and Iraqi Song Kang azole in vitro drug sensitivity test. Petri dishes and microtiter plate inoculum and placed in a 35 ° C incubator cultured for 24h to read the results. Spectrophotometric determination the itraconazole microtiter plate each well of the turbidity, and the control wells turbidity decreased by 50% of the drug concentration corresponding to the holes of the itraconazole minimal inhibitory concentration, measured by ruler affixed with fluorine size of the the itraconazole paper Petri dish inhibition zone, results accurate to the millimeter. Results: A total of 186 strains of Candida isolated, of which Candida albicans 125 (67.2%), tropical concept 29 strains (15.6%), 28 (15.1%), Candida glabrata, Candida krusei 3 (1.6%), Portugal Candida 1 (0.5%). Candida albicans, C. glabrata, of Candida tropicalis separation rate in the People's Hospital of Wuhan University, respectively, 68.3%, 10.0%, 20.0%; Zhongnan Hospital, Wuhan University, respectively, 80.9%, 4.2%, 14.9%; Tongji Hospital, Huazhong University of Science and Technology, respectively was 58.2%, 25.3% and 12.7%, respectively. 78 Candida albicans, 17 C. glabrata, 14 Candida tropicalis, 2 grams soft total of 111 experimental strains of Candida fluconazole sensitivity was 90.1%, of which Candida albicans is 97.4%, smooth candidiasis 58.8%, Candida tropicalis 100%; itraconazole sensitivity was 88.2% and 96.1% which Candida albicans, Candida glabrata 52.9%, Candida tropicalis 100%.
