Hepatocellular carcinoma (hepatocellular carcinoma, HCC) is the most common malignant tumor . Possible mechanism of primary liver cancer in rats model of hepatocellular carcinoma to establish the use of chemical inducers diethylnitrosamine (DEN) . The rat hepatoma and normal liver tissue from each of the three samples , samples from both proteomics and gene expression analysis . Proteomics technology using 2D-DIGE and MALID-TOF-MS technology . Gene expression profiling using Agilent 's latest rat 44K genome microarray analysis . Results: Proteomics: 2D-DIGE and MALID-TOF-MS proteomics technology we analyzed 18 protein differences between these points correspond to differences in protein expression of DEN - induced rat liver cancer model . In these tumor-associated protein , significantly up-regulated proteins, including GST superfamily , aldehyde - keto reductase superfamily and hypoxic stress related proteins corresponding members . Downward Corydalis acyl the acetoacetate enzyme , protein in liver cancer have been reported , and 3 - oxo - 5 -B steroids -4 lactate dehydrogenase , and its absence can cause liver dysfunction . Also found in rat hepatoma tissue dermcidin, a sweat gland - specific proteins were significantly increased . Proteomics conclusion is further proved by the western and immunohistochemistry . Corresponding sample through aligent chip analysis. Bioinformatics conduct clustering and functional classification analysis of the results show , the 789 genes significantly with upregulation of 559 genes significantly with lowered these significant changes in genes associated with cell cycle , cell infiltration and metastasis , cell signal transduction and cell metabolism , etc. closely related. These genes may be the use of chemical inducers of liver cancer markers in the early diagnosis of the tumor , looking for drug targets , play an important role in the prognosis , monitoring and individualized treatment . Chemical inducer of liver cancer model with human hepatocellular carcinoma have similar , so the data is important to study the mechanism of hepatocellular carcinoma . Our results suggest that the integrated use of proteomics and genomics technologies help us to identify the mechanisms of tumor-associated proteins and understanding of tumorigenesis .
