Abstract

Many degenerative diseases of the central nervous system is considered to be caused by the spontaneous apoptosis, cerebral ischemia and hypoxia injury is often caused by nerve cells the Ca 2 overloaded, causing the occurrence of neuronal apoptosis. In recent years, studies have shown that magnesium ions as an endogenous protective factor not only involved in important cellular metabolism and functional regulation of brain tissue, and have a definite protective effect on nerve cells. The study found that the brain damage brain tissue and nerve cells magnesium ion content, serum magnesium ion content were significantly decreased. Animal experiments have shown that supplemental magnesium ions reduce the experimental animal brain damage secondary to brain damage, improve neurological dysfunction, decrease in apoptosis of nerve cells. Animal experiments and epidemiological studies have shown that the lack of increase with age in older animals body the ubiquitous Mg 2 , Mg 2 absorption power will decline. Mg 2 lack of will affect the integrity of the cell membrane and its function, increased susceptibility to oxidative stress, which leads to cardiovascular disease, accelerate the aging process. Magnesium ion has a protective effect on neurons, mainly by inhibiting excessive activation of NMDA receptors play a role, the exact mechanism is unclear. Its protective role in neuronal apoptosis is also worthy of further study. The papers were established human neuroblastoma cells (SH-SY5Y cells) serum-free culture, H 2 O 2 processing and glutamate-treated cells model, study the Mg 2 of neuronal cells treated accordingly. Based on this, the experimental depth Mg 2 on the whole animal level, to study the old SD rat neurons. 1. Mg 2 serum-free culture cell model established into neuroblastoma cell line SH-SY5Y cells in serum-free culture cell model application MTT technology of Hochest33342 assay and quantitative PCR technology, morphology and gene mRNA levels on the effects of different concentrations of the Mg 2 serum-free culture of neuronal cells. The results showed that the serum-free culture decreased cell viability, apoptosis state NR2B mRNA expression levels decreased, to a certain extent after the join Mg 2 inhibit apoptosis. 2. Mg 2 H 2 O 2 processing cell model people into neuroblastoma cell line SH-SY5Y cells from oxidative stress model, application MTT, Hochest33342 method detection technology and quantitative PCR technology, from the effects of morphology and gene mRNA levels
