Abstract


Objective:Many investigate between IL-1B polymorphism and gastric cancer have been done on site -31 and -511 .In this study, we uite 1473 site to investigate the relationship betweenIL-1B polymorphism and gastric cancer. We also investigate the relationship between helicobacter Pylori infection and gastric cancer.Methods:1. Selection of population: 92 normal subjects and 63 gastric cancer patients.2. Measurement of interluekin-1B gene polymorphism: technique of gene chip was used, including DNA exteaction, PCR amplification;hybridization , showing colour , measurement and determining result.3. Measurement of helicobacter pylori : ELISA was used to measur the concentration of IgG , IgA, IgM of HP antigen in the blood serum .Result :1. IL-1B-31T/T genetype is a potential dangerous allele ,it increase the risk of susceptibility of gastric cancer.2. IL-1B-511C/C genetype is a potential dangerous allele ,it increase the risk of susceptibility of gastric cancer.3. IL-1B-1473G/G genetype is not a potential dangerous allele ,it cannot increase the risk of susceptibility of gastric cancer.4. In this study ,there is significant difference of frequency of IL-1B-3117T\ -511 C/C, between moderately and poorly-differentiated and well- differentiated group. It demonstrates that -31T/T\ -511 C/C are one of the risk factors which induce the poorly differertiation of gastric cancer.5. In the study ,there are nearly no linkage disequilibrium between two of this three genetype IL-1B-3U -51K -1473.6. In this study ,there is no significant difference of frequency of IL-1B-3K -511 -1473 between male and female of the patents and the controls.7. The infectious ratio of HP is significant higher in gastric cancer group than in the control group.lt shows that the infection of HP may induce gastric cancer.8. There is significant difference of frequency of IL-1B-511C/C genetype between the HP-negtive group and the HP-positive group .So IL-1B-511C/C genetype may induce gastric cancer.9. In the study ,it indicates that IL-1B allele is not associated with the lymph node metastasis in the gastric cancer.Conclusion: In a word , the susceptibility of gastric cancer is associated with IL-1B gene polymorphism and the infection of HP . Especialy , IL-1B-31T/T and -511 C/C may be used as indicators of susceptibility of gastric cancer and be used to alarm the susceptible individuals,prevent gastric cancer and estimate the prognosis of gastric cancer patients.
