Abstract

Obesity is an accumulation of body fat too many serious diseases affect human health. The common obesity is a polygenic disease or complicated disease, and well received by the genetic decision also affected by the environment, and between environmental and genetic factors and gene interactions may exist. Body mass index, fat content and fat percentage is an important risk factor for obesity, and there is a strong genetic determinants, genetic rate is generally higher than 0.5. A lot of research to these traits as obesity alternate representative type. Looking for disease genes for obesity have a large number of studies, including genome-wide scanning and candidate gene association studies. But the results are mostly inconsistencies. Studies have shown that imprinted genes or genome of the pathogenic mechanism of obesity affect about obesity genome-wide scan rarely consider the role of genomic imprinting, therefore, an object of this article is: in the large family of more than 4,000 samples full genome scan, and to consider the impact of genomic imprinting, to find obesity susceptibility genomic regions in the genome-wide level. The result of the scan of the whole genome that the chromosome 2q37 significant imprint linkage signal. The ordinary chain scan LOD score of 2.23, when the build maternal imprinting effect linkage scan chain signal increased to 3.34, the LOD score over the chain of this study is determined by a large number of computer simulations significant threshold. The studies also suggest to 2q31, 16q22, 3p14, 3q24 and 19q13 several chromosomal regions may contribute to the genetic formed of obesity. This study further prove the chain statistical genetic studies to consider the importance of genomic imprinting and phenotypic definition. Susceptible genomic regions by genome-wide linkage scan to find is an important aspect of the obesity of genetic research, direct study of candidate genes associated with obesity phenotype relationship is very important. LRP5 (low-density lipoprotein receptor-related protein 5) gene, a cell surface receptor-low density lipoprotein receptor family members, one of the LRP5 studies have demonstrated to play an important role in Wnt signaling pathway may control the bones and eyes formed at the same time has an important and significant role in glucose metabolism and cholesterol metabolism. Studies have found that LRP5 gene polymorphism is associated with some of the obesity-related complex diseases or traits. But still no relationship between LRP5 gene polymorphism with obesity
