Abstract


Background Biliary ischemia-reperfusion(I/R) injury is a common problem in many clinical conditions. It affects directly the recovery of the patients. Although many studies have been done in this field, it still remains an unsolved problem. More and more evidences indicate that the biliary tract is always one of the main targets of warm or cold I/R injury.It has been shown that biliary complications occurred in 7 to 30 percent liver transplantation, known as the most common complication, including biliary leak, biliary stricture, biliary infection, and so on. The biliary tract complication remains a challenging problem on liver transplantation, it may lead to death of the recipient. Lots of investigations confirmed that biliary I/R injury is closely associated with the outcome of the surgery. Thus, further study on the mechanism of biliary I/R injury is still an important task in this field.Now that no suitable animal model of biliary I/R injury is available and how long the liver can tolerance warm biliary ischemia has not yet been elucidated, we tried to establish a new animal model of biliary warm I/R in rabbits by clamping both common bile duct and common hepatic artery, and to investigate the tolerance limit of biliary blood inflow occlusion, as an experimental step in the approach to the clinical problems concerned.PartⅠEstablishment of the model of biliary warm ischemia-reperfusion in rabbits Methods Japanese rabbits, weighing 210-250g, were randomly divided into 5 groups(n=16 each). In the normal control group(SO), sham operation was performed on the common bile duct(CBD) and common hepatic artery(CHA). In the 2 hepatic artery occlusion(HAO2h, 3h) groups and the 2 combined CHA and CBD occlusion(HBO2h, 3h) groups, the warm ischemia lasted for 2h or 3h each.
