Effect of Hypothermia on the Early Stage of Intestine Mucosal Barrier Function after Traumatic and Hemorrhagic Shock

Abstract


Objective To observe the effect of varying hypothermia on inflammatory reaction, ischemical reperfusion injury and gut endotoxin transition by traumatic and hemorrhagic shock animal model. To investigate effect of hypothermia on intestinal mucous barrier function, and provide instruction for clinical therapy of trauma patients.Experiment methods Thirty-two New zealand rabbits were randomly divided into four groups: control group(groupⅠ,n=8), normal temperature group(groupⅡ,n=8), mild hypothermia group(groupⅢ,n=8), moderate hypothermia group(groupⅣ,n=8).Animals in groupⅠwere punctured carotid artery and vein. The right femurs of the animals in groupsⅡ,ⅢandⅣwere broken by a vise, and blood was drawn from the cervical vein after 30 minutes to establish traumatic and hemorrhagic shock models. The body temperature of the animals was maintained at normal level in groupⅡ, reduced to and maintained at 35-36℃and 32-33℃in groupⅢand in groupⅣrespectively. Blood samples were harvested in groupsⅡ,ⅢandⅣat 2,4,6,8 hours after trauma and in groupⅠat 2,4,6,8 hours after puncturing the carotid artery and vein respectively. Plasma endotoxin levels were estimated with tachypleus amebocyte lysate chromogenic. Experiment animals were sacrificed with exanguinating after harvesting blood sample, small intestine samples were collected for tissue homogenate. SOD activity, MDA and calcium ion levels in small intestine tissue homogenate were determined with chromatometry, Inflammatory cytokines and ICAM-1 levels were determined with ELISA. Pathology alteration were observed by electron microscope and light microscope.Results At 8 hours after trauma and hemorrhage: (1) TNFα,IL-6,IL-8, and IL-10 levels of intestine tissue homogenate were significantly higher in groupsⅡ,ⅢandⅣthan in groupⅠ(P<0.01); IL-6,IL-8 levels of intestine tissue homogenate were significantly lower in groupsⅢandⅣthan in groupⅡ; TNFαlevels of intestine tissue homogenate were significantly lower in groupⅢthan in groupⅡand lower than in
