ObjectiveType 1 diabetes is a disease due to autoimmune destroys of pancreatic β-cells. Current treatment is insulin injection, which can’t cure diabetes and not only bring suffering to patients, but also bring heavy burden to society and family. A new strategy is β-cell replacement therapy via islet transplantation. However, such an approach is limited by the scarcity of the transplantation material and the immunological rejection. Bone marrow mesenchymal stem cells are defined as non-hematopoietic adult stem cells capable of both self-renewal and multilineage differentiation. They have stable phenotypes and easy to isolate, culture, amplify in vitro. MSCs possess rapid proliferation ability and polytransdifferentiation potential in special culture condition. In order to study suitable cells-replacement source, bone marrow-derived cells from Kunming mice were separated, cultured and trans-differentiated into insulin-producing cells in this experiment. Materials and MethodsBM was obtained from the femurs and tibias of 4-6 weeks male Kunming mice and extruded by inserting a needle. At first, the BM cells were cultured in L-DMEM supplemented with 10% FBS for 1 hour. Then the BM cells were cultured with 1% dimethyl sulfoxide(DMSO) in serum-free DMEM for 3 days and then maintained for an additional 7 days in a DMSO-free H-DMEM medium containing 10% FBS.
