Abstract


Objective To research the exogenous gene transfection to human limbal stem cells (HLSCs) cultured in vitro and its profection on HLSCs injured by NaOH. In order to investigate the feasibility of the treatment for ocular surface diseases (OSD) at gene level. And to Provide preliminary experimental evidence for cure of OSD by genetic engineering combined with stem cell transplantation.The study includes three parts:ExperimentⅠ: Study on the proliferation,differentiation and assay of HLSCs in vitro.ExperimentⅡ: Study on the pEGFP-bFGF gene transfection to HLSCs and observation on its expression in cells.ExperimentⅢ: Study on the protecting effect of bFGF gene on HLSCs injured by NaOH.MethodsExperimentⅠ: Eyeballs were taken from the body within 6h after death and their superficial limbus were carefully denuded under the aseptic condition.
