Abstract

PrefaceCerebral ischemical reperfusion injury (CIRI) is the main risk factor that cause recogaition impairment. Hippocampus is critical for learning and memory in human and animals. Therefore the occurance of recognition impaiment is closely related with hippocampus ischemical reperfusion injury. There have been domestic reports stating that Aβplays a role in CIRI. And there are abroad reports claiming that Aβexpression is the result of neuron’ s acute reaction (to ischemia) and the following cascade reation leads to Aβlike change ( of neur-ous) . PSD -95 also participates in CIRI. In this study MCAO model was stab-lished with thread method and the expression of Aβ and PSD -95in ischemical reperfusion injured neurous of hippocampus was measured with immiobisto chemical methed in order to serve to provide information against CIRI.Materials and methods48 rato (2 ~ 3 months old, weighting 100 ~ 130g) from animal center of cau were randomly assigned into 3 groups; (1)Normal control group ( n = 6 ) ; (2)Sham operated group (n =6) ;(3)CIRI group(n =6). Establishment of CIRI model: The thread method of Longa was applied to the animals in CIRI group to create middle cerebral artery occlusion and reperfusion model. In CIRI group the rats’ brains were reperfused for 6h, 1d,2days,3days,7days respectively after 2hours ischemia. At the end of reperfusion, the rats were peritoneally anaesthesized with 10% chloral bydrate(0.3g/kg).4% poly formald chydet, was perfused to cerebral vessels through a catheter into left vertrical. Then the rats brains were take out for HE staining and Aβ and PSD - 95 immunohistachemical staining ( SABC
