Abstract


Zearalenone (Zearalenone, ZEA, F-2) is made from corn Fusarium graminearum Fusarium mycotoxins, the three lines sickle Jundeng fungi produced role of estrogen, mainly corn, wheat, barley, oats pollution and other crops and animal feed, reproductive and developmental toxicity, immunotoxicity, also have some impact on tumorigenesis, great health hazard to humans and animals. Yeast glucan is the main component of the yeast cell wall, immunomodulatory, anti-virus, adsorption of pathogens and their toxins, and other biological activity. In this paper, the nature of the beer yeast glucan and crosslinked carboxymethyl yeast glucan (CCMG) adsorption ZEA,. Firstly, the method of determination of ZEA content - high performance liquid chromatography. Trials to optimize the determination of conditions. The results show that this method is fast, good reproducibility and sensitivity and high recovery rate of 92.52% to 97.71%, the coefficient of variation (CV) of 1.14%. Soda, the alkali acid and enzyme soda of yeast from the beer extract alkali insoluble glucan product of qualitative and quantitative analysis, paper chromatography and infrared spectra show that the main products of β-D-glucan, and free mannose. The assay results showed that the three products ZEA: the enzyme Alkaline Extraction of β-D-glucan ZEA having a better absorption effect, the maximum adsorption amount reached 2.296μg/mg dextran. The enzyme Alkaline Extraction of yeast glucan as raw material, selection of epichlorohydrin as cross-linking agent, chloroacetic as carboxymethylation reagent, under the conditions of base-catalyzed synthesis CCMG. The degree of substitution (DS) test indicators, the preparation of the CCMG the better conditions determined by single factor and orthogonal experiments were as follows: reaction temperature of 50 ° C, the reaction time 5h, the the chloroacetic amount of 3.0g, NaOH concentration of 50%; under conditions resulting product substitution degree is about 0.5. The infrared spectrum showed: of CCMG the IR spectra in the wave number of 1600cm-1 strong-COO-ion characteristic absorption peaks, 1419CM-1 to the absorption peak of a methine group connected with the carboxyl, and show that it is crosslinked carboxymethyl glucan. Scanning electron microscopy, CCMG particles links into a group, damage to the structure of the particle surface, there is obvious damage grooves and deep caves, increasing the specific surface area. Adsorption tests showed that: good adsorption effect CCMG zearalenone The adsorption amount highest 18.643μg/mgCCMG higher than the adsorption amount of the yeast glucan.
