Abstract

Flavonoids are a class of polyphenols widely distributed in nature, with a wide range of biological and pharmacological activity. A large number of studies have shown that flavonoids scavenge free radicals, anti-oxidation, anti-mutation, anti-tumor, anti-bacterial, anti-viral, immune regulation, prevention and treatment of hardening of the arteries, lowering blood sugar, and other functions. With the development of the food industry and consumer attitudes change, health food natural active ingredients to become a modern pursuit of goals, flavonoids natural, highly active, quick and role of wide attentions gradually. The main content of this study are as follows: 1. Potassium permanganate - glyoxal - sulfuric acid chemiluminescence system Determination of flavonoids flavonoids common measurement methods: spectrophotometry, high performance liquid chromatography, capillary electrophoresis, ultra- supercritical fluid chromatography, TLC. The analysis of these methods are slow and expensive instruments. Chemiluminescent do not need any light source, to avoid interference from stray light, with a wide linear range, high sensitivity, simple equipment, easy to realize the advantages of automation and analysis speed. Through the optimization of the experimental conditions, the establishment of a potassium permanganate - glyoxal - sulfuric acid chemiluminescence system. The study found that the flavonoids in acidic conditions of the system has a strong sensitizing effect, whereby the establishment of a new method of potassium permanganate - glyoxal - sulfuric acid chemiluminescence determination of flavonoids. With this method, naringenin, naringin, baicalin, quercetin, puerarin, rutin, luteolin-7-O-half-galactosidase, icariin, dye lignin More than nine The Flavonoids the quantitative determination. The three kinds of chemiluminescence determination of antioxidant activity of the antioxidant capacity of flavonoids is the most important biological functions of flavonoids. The determination of the antioxidant capacity of flavonoids used method is visible absorption spectrometry using flow injection chemiluminescence few flow injection chemiluminescence antioxidant mechanism of flavonoids, and compare their activity, and to explore the flavonoids the relationship between the structure and antioxidant activity. (1) hydrogen peroxide cleared through the optimization of the experimental conditions, the establishment of the luminol - hydrogen peroxide - carbonate buffer chemiluminescence system. The study found that the system has a strong inhibitory effect of flavonoids and anthraquinone compounds under alkaline conditions, whereby the establishment of the anti-hydrogen peroxide determination of flavonoids and anthraquinone compounds. The method has been used for 18 flavonoids and antioxidant capacity of anthraquinone compounds measured and calculated their IC50 of. Experimental results show that luteolin, rutin, luteolin-7-O-half-galactoside, quercetin anti ability of hydrogen peroxide. (2) the removal of the hydroxyl radical through the optimization of the experimental conditions, the establishment of the luminol - hydrogen peroxide - copper sulfate - carbonate buffer chemiluminescence system. The study found that the system has a strong inhibitory effect of flavonoids and anthraquinone compounds under alkaline conditions, and accordingly, Determination of flavonoids and anthraquinone compounds for anti hydroxyl radical. The method has been used for 18 flavonoids and antioxidant capacity of anthraquinone compounds measured and calculated their IC50 of. Experimental results show that rutin, puerarin, genistein, baicalin, daidzein, kaempferol, luteolin, luteolin-7-O-half-galactoside, quercetin anti hydroxyl radical than strong. (3) removal of superoxide anion through the optimization of the experimental conditions, pyrogallol luminol - o - sodium hydroxide chemiluminescence system. The study found that the system has a strong inhibitory effect of flavonoids and anthraquinone compounds under alkaline conditions, pursuant to which the establishment of the anti-superoxide anion Determination of flavonoids and anthraquinone compounds. The method has been used for 18 flavonoids and antioxidant capacity of anthraquinone compounds measured and calculated their IC50 of. The experimental results show that the strongest, luteolin, rutin, genistein, luteolin-7-O-galactoside, quercetin superoxide anion scavenging ability. The above results show that: (1) on the B ring of the flavonoid compounds hydroxy more antioxidant capacity is stronger, when the 3 ', 4' there are o-dihydroxy, antioxidant activity is greatly improved. (2) The Flavonoids C ring 3-hydroxy change will affect the antioxidant effect. (3) 5 hydroxyl flavonoids can enhance its antioxidant activity. (4) in the metal ion-induced free radical generating test system, the three hydroxyl groups on the C ring of the flavonoid compounds can effectively enhance its antioxidant capacity, while on the C ring in the system of free radicals induced in the non-metallic ions 3 bit hydroxyl but contributed little. (5) Flavonoids C the ring 2,3 double bond is present and there and four carbonyl conjugate role of antioxidant activity have little impact. (6) In the different antioxidant system, the antioxidant capacity of flavonoids is not the same, they follow different mechanisms in different systems. The three kinds of chemiluminescence determination of people's daily drinking tea antioxidant capacity in the optimization good the three antioxidant system based on the study of health tea antioxidant capacity. Determined by extraction and extraction process, the determination of anti forty-six kinds of health tea hydrogen peroxide, hydroxyl radicals, superoxide anion capacity. The results showed that the three chemiluminescence antioxidant system can be rapid determination of the antioxidant capacity of the health tea, provide useful guidance for the screening of antioxidants from natural products.
