Abstract

Research Objective: The prevalence of apoptosis during cardiac development of the organism, the physiological and pathological processes of the body during exercise, disease development process, but there is not yet fully grasp the mechanism of apoptosis. Through the experiment, trying to by exhaustive exercise model, to explore feature SD rat cardiac myocyte apoptosis and apoptosis developments and changes in the law, to gain evidence of an association with exercise stress and apoptosis, in addition can also be further to investigate the mechanism of exercise-induced myocardial apoptosis laid the theoretical basis for the future ongoing monitoring studies of exercise training. Methods: 110 10-week-old male Sprague-Dawley rats were randomly divided into the quiet control group (n = 10), exhaustive exercise group (n = 50) and exhaustive exercise gavage Danshen injection group (n = 50), where exhaustive set of exhaustive exercise and orally Danshen injection group were randomly divided into 5 group (n = 10): instantly, 6 h after exercise, 24h, 48h, and 72h, the movement of three weeks, every movement The strength 5m/min, gradually increasing speed to 30m/min within 10min slope 0o to maintain the strength of this movement, until rats exhaustive. Three groups of rats in the resting state and movement immediately after exercise, 6h, 24h, 48h, and 72h were sacrificed, the heart of the organization system made of tissue homogenates, take the preparation of paraffin sections of rat myocardial tissue and TUNEL labeling 40 × 10 times fluorescence microscope to observe the changes in the structure of apoptotic cardiomyocytes. Experimental results: (1) In the quiet control group, occasionally sporadic cardiac myocyte apoptosis obvious apoptotic nuclei can be detected, repeated exhaustive exercise group, significant apoptosis index than the quiet group elevated, and the apoptotic index difference was significant (p lt; 0.05). (2) under the microscope, able to observe the cell morphology of apoptotic cardiomyocytes irregular changes, chromatin condensation nucleus to the nuclear membrane surrounding the integrity of the mitochondrial outer membrane destruction into the vacuole, cell membrane and nucleus The film structure is destroyed, etc.. Salvia fed group and non-fed group after exhausting exercises on the same point in time, with a significant difference between myocardial apoptosis, but both dynamically change certain similarity. (3) the exhaustive exercise group, the the rat myocardium SOD activity and quiet control group decreased a lot, the significance of the difference was significant (p lt; 0.05) Salvia SOD activity decreased fed group than exhaustive set of ease. (4) after exhaustive exercise, repeated exhaustive exercise group rat heart mitochondrial calcium content increased significantly (P lt; 0.05) than the quiet control group, significant difference between Salvia increase the calcium content of the fed group without exhaustive set of obvious. Conclusion: repeated exhaustive exercise can lead to myocardial apoptosis, apoptosis phenomenon generated, perhaps there is a greater relationship with the exhaustive exercise intensity and exercise duration. Possible mechanisms that cause myocardial apoptosis, perhaps because the-mitochondrial Ca 2 of accumulation, then the start due to apoptosis program. Danshen injection can reduce repeated exhaustive exercise-induced myocardial apoptosis and has a protective effect on the myocardium.
