Abstract
Brucella P39 cytoplasmic binding protein is a good T- cell antigen can induce strong delayed -type hypersensitivity , and produce large amounts of INF -γ , so that it is recognized as a DNA vaccine candidate molecules . The purified recombinant P39 for serological diagnosis of animal brucellosis is an excellent antigen . P39 protein after transformation strains inoculated animal , the animal will not be able to P39 immune response , the P39 protein can be used as molecular markers to determine whether the animal is a natural strains . Brucella Rev.1 gene encoding P39 protein was amplified by PCR , sequencing and cloned into the cloning vector pB ] LlescipLSKII ( ) to construct the recombinant vector pBp39 then a section of pBp39 encoding P39 1008bp fragment replaced kan resistant fragment constructed suicide the recombinant plasmid pBp39 -kan . Electricity conversion method pBp39-kan Ru Bulu coli for suicide plasmid due pBp39-kan , pre-designed Kan resistance fragment is homologous recombination sequences rescued in the side of each of the P39 gene arm homologous recombination in the form of a double exchange , so that led to the replacement of the P39 gene , thereby building became who rouxii bacteria molecular marker vaccine strains . Antigen P39 protein expression were detected infected with Brucella bacteria and new build molecular marker vaccine strain of animals , such as test results are negative , then the the animal artificial immune ; positive test results , it shows that the animals as a natural infection .
