Abstract


To explore Cistanche effect the growth and development of the pig embryo pig embryos as experimental materials, design three test: Cistanche water extract of the screening test for the early development of in vitro culture of pig embryos effect; Cistanche added at different times water extract of pig embryos early in vitro development; of Cistanche water extract of the impact on the the NO concentration content of the pig early in vitro development of embryos. To open up a new way to improve cultured in vitro embryo development rate and expand the scope of application of Chinese medicine. Test: NCSU-23 0.4% BSA base fluid for the control group, to different concentrations Cistanche water extract for the test group, after the initial screening of four concentration further filter out the best concentration. Test results: of Cistanche water extract of the best concentration of 50μg/ml, its cleavage and blastocyst rate were 85.12 ± 2.9% and 31 ± 3.0%, significantly higher than the 76.73 ± 3.2%, 24.89 ± 4.3 % (P lt; 0.05), the test between the groups without significant difference. The results show that: add basic medium Cistanche water extract can effectively promote embryonic growth and development in vitro. Test: the embryos at different developmental stages of the required nutrients better effect of the extract in order to clear Cistanche water added in embryonic development, during which period the test in three periods of embryonic development, adding Cistanche, compare where period added better. Test results: 2-4cell embryo culture add the Cistanche water extract, the the blastocyst rate reached 48.72 ± 6.6%, higher than that of the control group the blastocysts rate of 25.08 ± 5.4% between the two, there are very significant differences (P lt ; 0.01). Moreover, the ratio of in vitro maturation (IVM) and embryo culture (IVC) stage of the effect is better between the two significantly different (P lt; 0.05). The results show that: the embryo in vitro development of the ability to more effectively promote the 2-4cell add Cistanche embryo culture. Test: This test is based on the basis of preliminary studies on NO, NO content in culture to the blastocyst stage embryos were measured, observed to add Chinese medicine is an impact on embryonic NO content. Test results: After in vitro culture to the blastocyst stage, embryonic cells were higher NO content, the control group and the experimental group were 26.33 ± 1.90 and 20.30 ± 0.95. Add Cistanche water extract embryonic cells NO content was significantly reduced, with significant difference (P lt; 0.05) in the control group. The results showed that: Cistanche water extract is cleared to a certain extent with the free radicals produced by the embryo in vitro culture process, optimized biphasic regulatory role of NO in the cell signaling pathway, thereby reducing the formation of NO, increased embryonic The rate of in vitro development. These results suggest that: of Cistanche water extract of pig embryos in vitro growth and development effects significantly.
