Abstract

Vibriosis the three trituberculatus (portunus trituberculatus) is one of the high incidence in recent years the three trituberculatus breeding popular range wide, the most serious hazards disease. At present, the control of the disease is still based drug prevention and treatment as the main means, therefore, the urgent need to filter out the high efficiency, low toxicity, low-residue for three trituberculatus chemical drugs. Enrofloxacin (ENR) for third-generation quinolones, has low toxicity, broad spectrum antibiotic bacterial resistance mutation rates and other features, has been widely used in the livestock and aquatic animals, the prevention and treatment of bacterial diseases, is one of the few antimicrobial drugs approved for use in the aquaculture industry in China. Combined with the actual situation of the aquaculture production of this thesis to study the antibacterial effect of enrofloxacin the four kinds trituberculatus pathogenic Vibrio drug metabolism in normal and Vibrio alginolyticus infection three trituberculatus body to Enno The Sand Star to provide more scientific medicine in the prevention and control of aquatic animal diseases basis. The main findings are as follows: double dilution method the enrofloxacin of Vibrio alginolyticus (Vibrio alginolyticus), minimum vibrios (Vibrio mimicus), Harvey vibrios (Vibrio harveyi) and Vibrio vulnificus (Vibrio vulnificus) four common aquatic pathogens minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC), and on this basis, to study the of enrofloxacin different drugs concentration (0MIC, 1MIC, 2MIC, 4MIC) of the four bacteria bactericidal kinetics and antimicrobial effect (PAE). The results show that: enrofloxacin the Vibrio alginolyticus minimum vibrio, Vibrio harveyi and Vibrio vulnificus MIC were 0.8 mg / L and 0.2 mg / L and 0.4 mg / L and 0.2 mg / L Various concentrations of enrofloxacin are generated effects after varying degrees of antimicrobial enrofloxacin minimum Vibrio alginolyticus, Vibrio harveyi and Vibrio vulnificus in 1 MIC, 2 MIC and 4 MIC concentrations PAE were 0.25 h, 0 h, 0.07 h, 0.64 h; 0.37 H, 0.08 h, 0.86 h, 1.76 h; 0.83 H, 0.19 h, 1.09 h, 1.85h. The reversed-phase high-performance liquid chromatography (RP-HPLC) studied normal and Vibrio alginolyticus infection the three trituberculatus oral 10 mg / kg dose of enrofloxacin vivo pharmacokinetics law with 3P97 software drug when data on the organizations compartment model fitting. The results showed: enrofloxacin in normal and Vibrio alginolyticus infection Portunus trituberculatus hemolymph, muscle and hepatopancreas metabolic processes are in line with a two compartment open model, and its metabolite ciprofloxacin not space compartment model to describe. Enrofloxacin concentration after oral administration the normal three trituberculatus hepatopancreas, muscle and hemolymph organization in a rapid increase in about 1 h administration peak the C max , respectively 11.235μg / g, 0.850μg / g and 0.858μg / g, and V d / F 7.954 L. kg -1 , 10.367 L. kg -1 and 0.345 L. kg -1 . The t 1/2β , respectively, to 283.361 h h 47.869 and 17.681 h, AUC were 245.618μg. g -1 . h -1 , 24.753μg. g -1 . The h -1 and 20.111μg. g -1 . h -1 . Enrofloxacin three trituberculatus infection in vivo rapidly absorbed and widely distributed organization of penetrating. The main pharmacokinetic parameters in the three tissues: C max 0.710μg / g, 1.009μg / g and 3.474μg / g, V d 111.231 L . kg -1 , 7.514 L. kg -1 and 2.813 L. kg -1 , t β1 / 2 to 102.124 h, 57.661 h and 66.390 h, the AUC as μg. g -1 . h -1 , 39.632μg. g -1 . The h -1 and 146.727μg. g -1 . h -1 . Compared with the normal group, the infection three trituberculatus drug absorption and elimination rate accelerated significantly lower maximum plasma concentration and extent of absorption, the hemolymph apparent volume of distribution and muscle drug area under the curve increased significantly, hemolymph and muscle multiplet phenomenon, and the second peak is higher than the first peak. Vibrio alginolyticus infections cause the three trituberculatus loosely organized, increase in membrane permeability, the hepatopancreas tissue damage physiological change. Enrofloxacin metabolite ciprofloxacin could be detected in the tissues of normal and infected trituberculatus, but at low levels. Enrofloxacin maximum residue limits (MRL) in accordance with the Ministry of Agriculture animal food MRLs of veterinary drugs under 50μg/kg, infected The three trituberculatus oral enrofloxacin withdrawal period of not less than 450 degrees day, the water temperature was 25 ° C, the withdrawal period should be ≥ 18 d.
