Abstract

Polyacrylamide electrophoresis technique to study the genetic diversity of the three sea area of ??Haizhou Bay, Xiangshan Harbor, Liaoning, Liaodong Bay Clam groups. Were detected in the three groups of nine isozymes to 18 loci, three groups of polymorphic loci ratio (P .95 ) of 61.11%, 55.56%, 64.71% average heterozygosity observed value (Ho) range between 0.1689-0.1770, the effective number of alleles of loci were 1.4417,1.3917,1.4219, rich genetic diversity. The various groups widespread phenomenon of loss of heterozygosity, inbreeding coefficient (Fis) were 0.2140,0.1232,0.1797. Compared genetic differentiation (Fst) between the three groups and groups of every generation transport number (Nm), the results show that the the Haizhou Bay groups and Xiangshan Port groups genetic differentiation smaller Fst = 0.0448; Liaodong Bay population and the other two genetic differentiation between populations, Fst = 0.0755,0.0812. The cluster analysis results also show that The Haizhou Bay groups first Xiangshan Port groups together, after Liaodong Bay population clustering. In addition, we discuss the Clam population differentiation mechanism, the correlation coefficient of inbreeding and reproductive biology. Polyacrylamide gel electrophoresis analysis of of Ruditapes philippinarum groups of four sea area of ??Fujian Pingtan Island, Xiangshan Harbor, Haizhou Bay and Liaoning Liaodong Bay biochemical genetic characteristics. The results show that: the Philippines Ruditapes four groups of polymorphic loci ratio of 62.5% to 75.0%, with an average heterozygosity observations in the 0.2743 to 0.3387, close to the average expected heterozygosity, average effective number of alleles in the 1.4891 to 1.6325 between higher levels of genetic variation, loss of heterozygosity was not significant. Index of genetic differentiation among the comparison groups and groups of each generation transport number found genetic differentiation among the three groups of Pingtan Island, Xiangshan Port and Haizhou Bay no significant genetic differences between the groups, with three groups of Liaodong Bay. Cluster analysis is the same: Pingtan Island groups first Xiangshan Port group clustering, and then meet with the Haizhou Bay groups last phase exchange Liaodong Bay population. Preliminary discussions heterozygous deletion causes and groups was not significant genetic differentiation mechanism. PCR amplification collected from Lianyungang, Ningbo, and Jinzhou three sea areas the Clam (S.subcrenata) groups ribosomal genes transcribed spacer ITS-2 sequence has been a 500bp clear bands, purified and directly sequenced. ITS-2 sequence of bases in Clam T, C, A, G four bases, the average content of 26.1%, 24.3%, 27.3%, 22.4% GC content lower than AT content. The MEGA3 Software detected a total of 27 variable sites, of which there are three nucleotide insertion / deletion mutation sites, 24 conversion or transversion nucleotide sites of 27 haplotypes. Groups genetic distance in between 0.000-0.024. NJ MP molecular phylogenetic tree constructed using ITS-2 sequence show no obvious clustering of groups, no obvious genetic differentiation between different geographical groups Clam. Clam, Kui of cockles (S.broughtonii) and mud cockles (T.granosa) 3 Arcidae ITS-1 and ITS-2 sequences were amplified PCR products were purified T vector cloning and sequencing. ITS-1 and ITS-2 sequence, respectively application ortho to the NJ and UPGMA molecular phylogenetic tree constructed display: Clam and Scapharca, First clustering the connected method and the unweighted arithmetic average group, and then together with mud cockles. This will Clam and Scapharca, vesting with 佩尔森纳贝 class classification Clam case, mud cockles vest to mud cockles Zodiac consistent.
