Abstract


Objective: ① quantitative detection of gastric mucosal benign biopsy and gastric carcinoma tumor-associated macrophages (TAMs) and micro-lymphatic vessel density (LMVD). (2) analysis of changes in the distribution and number of benign gastric mucosal biopsies and gastric carcinoma tumor associated macrophages. (3) analyze the the benign gastric mucosal biopsies and gastric carcinoma LMVD the distribution and the number of changes. The ④ analysis of gastric carcinoma tumor-associated macrophage infiltration and lymphangiogenesis (LMVD). The ⑤ analysis of gastric carcinoma tumor-associated macrophage infiltration and lymphangiogenesis (LMVD) and clinical pathologic parameters. And ⑥ explore the tumor associated macrophages in the the gastric lymphatic formation of lymph node metastasis. Methods: First Hospital of Lanzhou University, from August 2003 to June 2005 surgical resection archived paraffin-embedded specimens of 68 patients with gastric cancer and 15 cases of gastric mucosal benign biopsy specimens. Gastric cancer specimens divided into Group A: 30 cases of differentiated carcinoma (papillary adenocarcinoma and tubular adenocarcinoma), group B: undifferentiated carcinoma (poorly differentiated adenocarcinoma and signet ring cell carcinoma) 38 cases of group C: T1 T 2 32 patients, group D: T 3 T 4 36 cases, E group: 38 cases with lymph node metastasis, Group F: 30 cases of patients without lymph node metastasis, Group G: Ⅰ Ⅱ 27 cases, H: Ⅲ Ⅳ 41. Did not receive preoperative chemotherapy and other adjuvant therapy. Formalin-fixed, paraffin-embedded specimens were serially sectioned slice thickness of 5 μm. Immunohistochemical S-P method of CD68 marker macrophages, VEGFR-3 tag lymphatic microvessel light microscope counting Weidner method macrophages and lymphatic microvessel density. The experimental data in the statistical software SPSS10.0 t test, Pearson correlation analysis. Results: macrophage counts and LMVD in gastric cancer tissues (74.062 ± 23.299,8.185 ± 3.265) was significantly higher than benign gastric mucosal biopsy tissue (58.253 ± 18.683,5.853 ± 2.357), the difference was statistically significant (P = 0.016,0.011) . Macrophages count and LMVD of lymph node metastasis and TNM stage were significantly correlated: the macrophage count LMVD in lymph node metastasis group (80.605 ± 19.765,9.363 ± 3.253) was significantly higher than that without lymph node metastasis (65.773 ± 25.070,6.700 ± 2.663 ), the difference was significant (P = 0.008,0.001): Ⅲ Ⅳ group (78.737 ± 21.484,8.995 ± 3.403) was significantly higher than Ⅰ Ⅱ group (66.963 ± 24.534,6.963 ± 2.671), the difference was statistically significant (p = 0.041 , 0.011). Also significantly associated macrophage count and LMVD gastric tissue differentiation and depth of invasion can not be determined in undifferentiated group (poorly differentiated adenocarcinoma and signet ring cell carcinoma) (76.332 ± 23.450,8.821 ± 3.323) than in differentiated group (papillary adenocarcinoma and tubular adenocarcinoma) (71.187 ± 23.177,7.387 ± 3.068), but the difference was not significant (P = 0.370,0.072); T3 T4 group (78.789 ± 23.886,8.761 ± 3.541) higher than T1 T2 (68.744 ± 21.769,7.544 ± 2.851), the difference was not significant (P = 0.076,0.126). Conclusion: TAMs count and LMVD in gastric cancer tissues was significantly higher than that of benign biopsies of the gastric mucosa and lymph node metastasis and TNM stage was significantly associated, prompt gastric tissue infiltration of TAMs may promote tumor-associated lymphatic vessel formation and thus promote lymph node metastasis may gastric cancer lymph node metastasis.
