Background and purpose of cervical cancer is a common gynecological malignancy, surgery and radiotherapy is the main treatment. Despite improved surgical techniques, radiotherapy equipment and technology continues to improve, cervical cancer overall 5-year survival rate has not improved significantly, still hovering around 50%; due prognostic mainly due to the transfer of cervical cancer, so how to improve the limited The therapeutic effect of the nature of advanced cervical cancer is the focus of the present study. Invasion and metastasis of malignant tumors of the basic biological characteristics. Tumor metastasis is a highly selective, multi-step, continuous process of interaction of multiple factors involved in the tumor cells and host cells, require further changes in gene expression in tumorigenicity, such as metastasis suppressor gene loss of expression and matrix high expression of metalloproteinase (matrix metalloproteinases, MMPs). KiSS-1 gene is a newly discovered metastasis suppressor gene expression by inhibition of cell mobility, promote apoptosis, inhibition of tumor cell proliferation and negative regulation of matrix metalloproteinase the -9 (matrix metalloproteinases-9, MMP-9) of and so to inhibit tumor metastasis. KiSS-1 gene is associated with a variety of malignant tumors of the transfer and provide prognostic information. MMP-9 is the largest molecular weight enzyme MMPs, expressed in a variety of malignant tumors, tumor invasion and metastasis. At home and abroad, yet metastasis of KiSS-1 gene with cervical cancer, there is no reported interaction of KiSS-1 and MMP-9 in cervical cancer. This study analyzed of KiSS-1, MMP-9 transfer relationship with cervical squamous cell carcinoma, as well as KiSS-1 and MMP-9 in cervical squamous cell interactions, designed to provide clinically effective diagnostic predictors and new therapeutic target. Method 1. Collection of normal cervical epithelial tissue of 12 cases, 46 cases of CIN cases and 14 cervical squamous cell carcinoma, KiSS-1 mRNA expression was detected by in situ hybridization analysis of the relationship between the KiSS-1 mRNA expression in cervical squamous cell carcinoma clinicopathological factors, KiSS-1 mRNA expression and protein metastin. 2. Immunohistochemical detection of metastin and MMP-9 protein expression in the three organizations, the analysis of the relationship between metastin and MMP-9 protein expression and cervical squamous cell carcinoma clinicopathological factors, as well as metastin and MMP-9 protein expression correlation. 3. Statistical analysis: Data SPSS13.0 statistical software for analysis. X ~ 2 test (α correction) and Fisher's exact comparison between the sample rate, both correlation test using Spearman rank correlation analysis, taking α = 0.05 is significant with test standards. Results 1. The positive expression rate of KiSS-1 mRNA in normal cervical epithelial tissue, CIN and cervical carcinoma were 100% (12/12), 85.7% (12/14) and 32.6% (15/46), the differences among the three groups statistically significant (P <0.01); positive expression rate of cervical squamous cell carcinoma group was significantly lower than the CIN group and the normal group, the difference was statistically significant (P <0.01), no statistically significant difference between the CIN group and normal group (P> 0.05). Loss of expression of KiSS-1 mRNA and the clinical stage of cervical squamous cell carcinoma and lymph node metastasis, histological grade (P> 0.05) (P <0.01, P <0.05), and the patient's age. 2. the metastin normal cervical epithelial tissue, CIN and cervical squamous cell carcinoma, the positive expression rate of 100% (12/12), 78.6% (11/14) and 39.1% (18/46), the differences among the three groups was statistically significance (P <0.01); positive expression rate of cervical squamous cell carcinoma group was significantly lower than the CIN group and the normal group, the difference was statistically significant (P <0.05, P <0.01) between the CIN group and the normal group, the difference was not statistically significance (P> 0.05). of metastin the loss of expression of cervical squamous cell carcinoma clinical stage and lymph node metastasis, histological grade (P> 0.05) (P <0.05), and the patient's age. 3. MMP-9 protein in normal cervical epithelial tissue, CIN and cervical squamous cell carcinoma, the positive expression rate was 8.3% (1/12), 64.3% (9/14) and 71.7% (33/46), the differences among the three groups was statistically significant (P <0.01); positive expression rate of cervical squamous cell carcinoma and CIN group with normal group there was significant difference (P <0.01, P <0.05), the positive expression rate of cervical squamous cell carcinoma and CIN group was no significant difference (P> 0.05). MMP-9 protein highly expressed in cervical squamous cell carcinoma lymph node metastasis related (P <0.05), and the patient's age, histological grade and clinical stage (P> 0.05). 4. Cervical tissue KiSS-1 mRNA and metastin expression was positively correlated (r_s = 0.664, P <0.01), metastin in cervical squamous cell carcinoma, and MMP-9 protein expression was negatively correlated (r_s = -0.387, P <0.01). Conclusion 1. KiSS-1 mRNA and metastin expression in normal cervical epithelium and CIN in high loss of expression in cervical squamous cell carcinoma; KiSS-1 mRNA and metastin the loss of expression of the clinical stage of cervical squamous cell carcinoma and lymph node metastasis, suggesting that KiSS-1 may play an important role in the invasion and metastasis of cervical squamous cell carcinoma. 2. In normal cervical epithelium loss MMP-9 expression in cervical squamous cell carcinoma and CIN in high expression; MMP-9 expression in cervical squamous cell carcinoma lymph node metastasis, suggesting that MMP-9 may be involved in the invasion and metastasis of cervical squamous cell carcinoma. 3. The cervical tissue KiSS-1 mRNA expression and metstin were positively correlated, suggesting that the transcription and expression of the gene may have consistency. Metastin and MMP-9 protein expression in squamous cell carcinoma of the cervix were negatively correlated, suggesting that the two may exist in the invasion and metastasis of cervical squamous cell interactions.
