The EliVisionTM Plus immunohistochemical technique and in situ hybridization technique were used to detect Snail, E-cadherin, CD44v6protein and mRNA in chronic cervicitis, cervical intraepithelial neoplasia (CIN), and cervical cancer. This study aimed to investigate the aspects below:Effects of methods for different assay kit and antigen repair on Snail expression; To investigate the expression of Snail, E-cadherin, CD44v6protein and gene in Cervical cancer and its relationship with clinicopathological features; The relationship between Snail and E-cadherin or CD44v6.The samples of tumors were taken from70cases of cervical cancer tissues,35cases cervical intraepithelial neoplasia(CIN) and15cases of chronic cervicitis, which were admitted to the251st Hospital of PLA from2008to2010. The paraffin block of the largest size was taken, the sections in4μm thickness were kept in oven at65~70℃for1~1.5hours and the expression of Snail、E-cadherin、CD44v6protein and gene was detected by streptavidin-peroxidase immunohistochemistry combined in situ hybridization technical inspection in these samples. Selected dilute/concentrated Snail monoclonal antibody as immunohistochemical markers in70cases of cervical cancer, and the concentrated Snail monoclonal antibody was diluted by1:200.The expression rate of the concentrated Snail was much higher than that of the dilute Snail, and some cases which were negatively stained by the dilute Snail were positive by the concentrated Snail. There was statistically significant difference between the two groups (χ2=4.145, P<0.05). The positive rates of Snail protein and gene were(26.7%、20.0%)、(65.7%、60.0%)、(87.1%、80.0%) in chronic cervicitis、cervical intraepithelial neoplasia(CIN) and cervical cancer respectively; The expression of Snail among groups had statistical difference (P<0.05). There was no statistical difference between Snail protein and gene expression. The expression of Snail protein and gene were related to lymph node metastasis (P<0.05; P<0.05), but not to age, differentiation of cancer tissues, FIGO stage and invasion depth (P>0.05). The positive rates of E-cadherin protein and gene were(93.3%、93.3%、(54.3%、48.6%)、(30.0%、21.4%) in chronic cervicitis, cervical intraepithelial neoplasia(CIN) and cervical cancer respectively; The expression of E-cadherin among groups had statistical difference (P<0.05). The expressions of E-cadherin protein and gene had no statistical difference (P>0.05). The expression of E-cadherin protein and gene were related to lymph node metastasis (P<0.05; P<0.05), but not to age, differentiation of cancer tissues, FIGO stage and invasion depth (P>0.05). The positive rates of CD44v6protein and gene were(20.0%、13.3%)、(48.6%、42.9%)、(84.3%、78.6%) in chronic cervicitis, cervical intraepithelial neoplasia(CIN) and cervical cancer respectively; The expression of CD44v6among groups had statistical difference (P<0.05). The expression of CD44v6protein and gene were related to lymph node metastasis and FIGO stage (P<0.05, P<0.05),(P<0.05, P<0.05), but not to age, differentiation of cancer tissues, and invasion depth (P>0.05). The expressions of Snail, E-cadherin protein and gene in cervical cancer showed negative relationship(r=-0.400, P<0.01;r=-0.783, P<0.01). The expressions of Snail, CD44v6protein and gene in cervical cancer showed positive relative relationship (r=0.420, P<0.01; r=0.348, P<0.01). The expressions of E-cadherin CD44v6protein and gene in cervical cancer showed negative relationship(r=-0.660, P<0.01;r=-0.745, P<0.01).The following conclusions were got through the above expressions: The expression of Snail in cervical cancer existed false negative. Therefore, concentrated Snail monoclonal antibody was suggested to use. There was not statistical significance between Snail protein and gene expression, between E-cadherin protein and gene expression or between CD44v6protein and gene expression respectively, which demonstrated that there was no difference between those protein and gene expression. Snail, E-cadherin and CD44v6were involved in the development and progress of cervical cancer, and were also related to lymph node metastasis, and were thought as a prognostic marker of cervical cancer. The negative correlation between Snail and E-cadherin and the positive correlation between Snail and CD44v6further demonstrated that they play an important role in cervical cancer cell proliferation and metastasis.
