Kidney cancer is a common malignant tumors of the urinary system, and its incidence has been increasing trend, a serious threat to human health. Renal cell carcinoma, the molecular mechanisms of development is a hot research topic in recent years. Which cytokine signaling concern. Cytokines are decisive factors to adjust the cellular differentiation and proliferation, these molecules are mainly cytokine-induced JAK / signal transducer and activator of transcription for (signaltransducer and activator of transcription, STAT) signal pathway play a biological role. The occurrence of abnormal and malignant tumors of the JAK / STAT pathway. Moreover, cytokine signal transduction inhibitor (suppressor of cytokinesignaling and SOCS in) family regulate cell growth, differentiation play an important role. The main mechanism for the inhibition of JAK / STAT signaling pathway play a biological role. The SOCS family's role in the process of malignant disease has been a cause of concern. Liver cancer, pancreatic cancer, stomach cancer, myeloma, lung cancer and breast cancer research reported have confirmed SOCS gene expression in malignant missing. And found abnormal height of SOCS gene promoter CpG island methylation is the main mechanism of gene silencing. So far has not been about SOCS role in renal cell research reported SOCS-3 gene in this study was determined by RT-PCR method. The level of renal cell carcinoma, and SOCS-3 methylation status by methylation-specific PCR detection. SOCS-3 in renal cell carcinoma, in order to explore the development. In this study, we apply the RT-PCR method determination of 10 human cases of normal kidney tissue, 15 cases of renal cell carcinoma, SOCS-3 gene expression 15 cases paracancerous organizations, and detected by methylation-specific PCR SOCS -3 methylation status. Detected compared with normal kidney tissue, detected in 86.7% of renal cell carcinoma the SOCS-3mRNA expression levels tended to decrease (compared with normal control tissue, p <0.05); cancer tissue and normal kidney tissue were detected SOCS- 3 expression, the expression levels of the two no significant difference (p> 0.05). Methylation specific PCR results showed that: 13 cases (86.7%) with primary renal cell carcinoma specimens SOCS-3 promoter hypermethylation, and consistent with the reduction or absence of expression of SOCS-3. The results of this study suggest that the RCC common SOCS-3 expression is missing or reduced; often occurs SOCS-3 promoter hypermethylation and SOCS-3 expression silence.
