A Proteomic Analysis of Aorta from Spontaneously Hypertensive Rat

Abstract


The pathological basis of hypertension is vascular remodeling, however, the mechanism of vascular remodeling during hypertension remains unclear. The differentially expressed proteins in aortas from spontaneously hypertensive rats (SHR) and their normtensive counterpart, Wistar Kyoto rats(WKY) were examined by using two-dimensional electrophoresis(2DE) technique.Within pH range of 3 to 10, 2000 polypeptides were separated per gel. After analyzing gels 50 differentiated expressed proteins were found, among which 27 were highly or only expressed in SHR and 23 in WKY. After MALDI-TOF-MS analysis, 20 proteins were identified, among which 9 had highly confident ID, including Rho GDP dissociation inhibitor alpha (RhoGDI alpha).RhoGDI alpha was verified at mRNA and protein level among 4, 12 and 18 week-old age groups of SHR and WKY. RhoGDIαwas significantly highly expressed in 18 week-old group of SHR. Meanwhile, when the vascular smooth muscle cells (VSMCs) of SHR and WKY were treated with angiotensin II and angiotensin II type I receptor(AT1-R) antagonist (L158809) respectively, RhoGDIαwas upregulated by angiotensin II and downregulated by L158809, which suggested that the expression of RhoGDIαmay be enhanced through angiotensin II-AT1R pathway. These results provided a proteomic angle to find the vascular remodeling related protein, RhoGDIα, during hypertension and some data for the mechanism of vascular remodeling during hypertension.
