Non - transmembrane protein tyrosine phosphatase SHP-2 is a kind of exist in mammals , and it plays an important role in the regulation of cell biological variety of signal transduction and control of cell activity . For the study of the biological function of SHP-2 , we choose nematodes (Caenorhabditis elegans) as a model organism , RNA interference PTP-2 ( nematodes in SHP-2) of the function . First, we designed synthetic primers , a PTP- 2 gene was amplified using PCR method from the cDNA library of the nematode , and cloned into the expression vector pGEX- 2T , and efficient expression in E-coli BL21codonplus pLysS ; Secondly, we application of preparative electrophoresis to obtain a high purity of the GST-PTP-2 fusion protein , and the resulting fusion protein-immunized rabbits to obtain a higher specificity of PTP - 2 polyclonal antibody ; Finally, on the basis of the above work , we application of RNAi technology interference ptp-2 gene expression in wild-type C. elegans nematode biology phenotype , we found that the interference group, a significant difference with control group C. elegans nematode aspects of life as well as non - normal death , indicating that PTP- muscular system development process played an important role in the regulation of nematode vagina , also involved in the life related regulation of signal transduction pathways .
