Abstract


Objective: To establish D-galactosamine-induced acute liver failure animal model, its application of N-acetylcysteine ??(N-Acetylcysteine, NAC) treatment, observation NAC treatment effects, in-depth study of the mechanism of the drug. Methods: Chinese Experimental Mini Pig, female, 12, were randomly divided into two groups: control group (n = 6) and treatment group (n = 6). The application of D-galactosamine 1.2g/Kg intravenous induction of acute liver failure animal model. 6 hours after administration of the treatment group, 24 hours, 48 ??hours, were given NAC150mg/kg, 50mg/kg, 100mg/kg adding to 100ml intravenous infusion of 5% glucose solution for each subsequent 24 hour intervals given NAC100mg/kg adding 5% glucose solution 100ml intravenous infusion. 6 hours after the administration of the control group, and after 24 hours every 24 hours, respectively, intravenous infusion of 5% glucose solution 100ml. Observe and compare the two groups of the general condition of the animals, survival time, physiological and biochemical indicators of intracranial pressure, pathological changes and serological factors change. Results: the animals of the control group, the mean survival time was 64.7 ± 8.7 hours, and died of severe liver failure. Treatment group, survival time was 66.2 ± 10.6 hours, The mean survival time, the difference was not significant (P = 0.73, P> 0.05). Compared with the control group, the treatment group, a number of biochemical markers, intracranial pressure, inflammatory cytokines have different degrees of improvement (p <0.05 ~ p <0.01). Pathological examination of the treatment group in addition to the 60 hour group, the rest of the time points Histopathology showed better recovery than the control group. Immunohistochemical examination, the control group, PCNA positive expression, but a smaller number, slightly stained; treatment group, the number of positive expression, the coloring is relatively deep. Two sets of positive cell count after statistical analysis, the two sets of positive expression rate difference was significant (p <0.05); Caspase-3 positive expression in both the control group and the treatment group, the cytoplasm and the nucleus are brown particles shape, density ranging from less than the control group, but the treatment group, the number of positive particles. CONCLUSION: NAC used in the treatment of acute liver failure have a certain effect to: ① NAC treatment can improve the body's general condition and function, to some extent, provide useful help for the recovery of liver function. ② The NAC treatment can reduce the body of TNF-a, of IL-1β inflammation cell factor of production, inhibit the rapid development of the inflammatory response and stability of the body of the inner environment, alleviate liver damage, liver cell regeneration, repair provide the conditions; ③ The NAC treatment to maintain the oxidation play a certain role and antioxidant balance by increasing the antioxidants SOD, reduced glutathione and nitric oxide content to remove oxygen free radicals produced by the liver oxidant, reducing oxygen free radicals damage the liver, since to protect the liver; ④ NAC can promote the regeneration of liver cells, is conducive to the liver function recovery. In summary, the application of N-acetylcysteine ??provides an effective way and a new choice has important clinical significance for the clinical treatment of liver failure.
