Abstract


[Objective] To study the feasibility of the application pseudomembranous fibroblasts to build artificial joint osteolysis in nude mice model . 【Method】 In this study, completed in May 2006 to January 2007 in the central laboratory and animal laboratory of Shanghai Sixth People's Hospital , 15 nude mice (SCID mice ) were randomly divided into three groups , each group 5 ; respectively , for the pseudomembrane fibroblast group (group I ) , the joint capsule fibroblast group (Group II ) , control group (Group III ) , pseudomembranous fibroblasts group from aseptic loosening of artificial joint revision surgery the the pseudomembrane organization , made ??fibroblasts isolated after 3 to 4 times passaged , the use of bone cement stimulate fibroblasts implanted in nude mice calvaria ; joint capsule fibroblasts group from the initial artificial joint replacement surgery remove the part of the joint capsule , isolated fibroblasts implanted in nude mice calvaria after with the same group I , the control group was sham-operated group ; after two weeks , the mice were sacrificed , remove the calvaria bone decalcification slices after treatment , the application of image analysis software osteolysis nude mice cranial suture quantitative analysis osteolysis of the area to determine the extent of bone resorption . [Results ] of pseudomembranous organizations and joint capsule of isolated cells were identified expression of fibroblast -associated antigen ( proline 4 - hydroxylase ) , pseudomembrane fibroblasts detected by RT-PCR found high expression of RANKL and of OPG mRNA , pseudomembranous tissue fibroblasts implanted nude mouse calvaria from the slice of the HE staining observed in nude mouse calvaria after measurement pseudomembrane fibroblasts group mean bone resorption through IPP software analysis The absorption area of 0.23 ± 0.030 mm2 (P lt; 0.05), significantly higher than the joint capsule fibroblast group (group Ⅱ ) ( 0.083 ± 0.013mm2 ) and control group ( group Ⅲ ) ( 0.078 ± 0.009 mm2 ) . The operability Conclusion fibroblasts isolated from pseudomembranous pseudomembranous into fiber cells implanted in nude mice can be successfully induced osteolysis formation , repeatability , and can be used as animal models to study artificial periarticular osteolysis .
