Abstract

Osteoporosis (OP) has already been a serious disease that threatens human health worldwide. The incidence of the disease has taken the seventh place among the normal diseases in the world.Puerariae radix (PR) is an exterior-resolving herb in Traditional Chinese Medicine (TCM). Sample of PR (TIP) has proved effects on quicken the blood and transform stasis in last several decades. PR is widely used in hypertension, coronary disease, myocardial infarction, etc. In recent years, some researches showed that TIP had good curative effect on OP in animal trials and cell trials.According to the clinical observation, TCM“BXW”can be used on the treatment of OP.Presently, there are little Chinese medicines available which have good curative effects on OP. Following the regulations of the development of new medicine, the studies of PR and Fufang on pharmacy and pharmacology had been done as following. In new drug exploitation, this experiment made pharmacy and pharmacological study on the new drug above typedⅣ(PR) andⅥ(Fufang).1. Measurement of total isoflavones and puerarin in TIPPuerarin is the control sample in determining the total isoflavones in TIP with ultraviolet spectroscopy (UV). The result of the total isoflavones in TIP is 70.14%.Puerarin is the control sample in determining the content of total puerarin in TIP with high performance liquid chromatography (HPLC). The result of the content of puerarin in TIP is 38.51%.2. Protective study on the effect of TIP on osteoporosis in ovariotomied rat.77 female Wister rats, 3-month old, were ovariotomied bilateraliy under anesthesia. 5 days later, animals were orally administrated with 80mg/kg.d (TIP-80), 40mg/kg.d (TIP-40), 10mg/kg.d (TIP-10), Nilestriol Tablets (E) or XianLing GuBao Jiaonang (ZY) respectively for 3 months. The Sham and OVX group were orally administrated with distilled water. The femoral BMC, BMD, strength and other parameters such as the content of Ca, P in tibia and serum, were detected after 3 months. Compared with OVX group, BMC, BMD, strength, content of Ca, P in tibia and serum were enhanced in TIP-40. The results indicated that TIP has protective effect on ovariotomied rats.3. Study on the pharmacy of TCM“BXW”The optimum technology on extraction of herbs F1 and F2 was 15 times of water, 3 times extraction and 1.5h each time; F3, F4 was extracted by 9 times of ethanol (60%) for 1 time, 1h each time. The preparation technology was that the portion of dried extract and dextrine was 1:2-2.5), and the contraction of ethanol was about 50% .4. Study on quality standardTLC and HPLC methods had been employed for discriminate and measure of valid component in TCM“BXW”. The method were feasible and easy. The quality standard of TCM“BXW”had been established.
