Abstract


Tumor heterogeneity of malignant cell populations , wherein karyotype heterogeneity is an important characteristic of tumor cells , but also promote tumor cell continuing evolution and resistant foundation . The heterogeneity of the karyotype instability leads to tumor cell chromosome . Chromosomal instability as a feature of the tumor has been widely recognized , but the chromosomal instability of the origin of ways as well as the instability of the aneuploid cell fate is still unclear . We will real - time photographic technique of the long process of living cells with fluorescence in situ hybridization combined , the first real-time photographic observation of living cells expressing green fluorescent protein and histone 2B fusion protein in HeLa cell division , and then determined by fluorescence in situ hybridization these chromosomes , cell division and chromosome segregation combined and then come to the answer to this question . The results show that the HeLa in abnormal number of chromosomes in the cells from more split , the most important is the the monocytes three-pole split and dual-core cell quadrupole splitting . More frequent partial fusion of the pole of daughter cells or full integration of dual-core cells or multinucleated cells . Fusion cells ( dual-core and multi-core cells) in the subsequent culture of death or the next round of multi-polar split . Only the unfused cells ( monocytes ) before normal bipolar division, so as to be able to stable genetic material is passed from one generation to the next generation , and therefore has the possibility of long-term survival . We further by single-cell colony formation test found that part of the multi- the very dividing cells can grow into a clone . In summary, the abnormal number of chromosomes in the cells divide to produce a multi-polar and escape the the fusion fate of the daughter cells will grow into a clone , suggesting that HeLa cells divide to produce a multi-polar genetic diversity , thus contributing to the HeLa cell chromosomal instability . These results indicate that aneuploid cells from split , they have the ability to continue to divide , and thus could be important for cancer .
