Abstract


Background and Purpose Primary liver cancer is one of common malignant tumors, hepatocellular carcinoma (hepatocellular carcinoma, HCC) accounted for 91.5% of primary liver cancer, primary liver cancer mortality rates for men and women respectively in the first malignancy third and fourth place. Tumorigenesis is a long-term, progressive, multi-stage, complex multi-step process, and abnormal cell proliferation and apoptosis regulation disorder is a common feature of all malignant tumors. Livin protein as inhibitor of apoptosis proteins (inhibitor of apoptosis protein, IAP) IAP family proteins was found, and its unique structure, able to protect cells against a series of specific allergen-induced stimulation, inhibition of apoptosis, therefore Livin gene is considered to be a new anti-apoptotic oncogene. Studies have shown that with a variety of tumor malignancy and prognosis, such as lung cancer, bladder cancer, stomach cancer, nasopharyngeal cancer. Livin in HCC related research at home and abroad no relevant reports in the literature. β-catenin is an important intracellular protein, which has a dual function, first-mediated cell adhesion of E cadherin important structural elements, and its abnormal changes in tumor invasion and metastasis processes play an important role: the second is WNT signaling key signal transduction pathway molecule, resulting in degradation of cytosolic free of obstacles β-catenin accumulation and the T cell factor / lymphocyte enhancer factor (TCF / LEF) incorporated into the nucleus, activating downstream target genes c-myc transcription , causing uncontrolled cell proliferation and differentiation, leading to tumorigenesis. HCC in the β-catenin gene mutations and protein overexpression in tumor incidence, prognosis, now the gene expression and tumor grade and prognosis of many controversies. This study immunohistochemistry Livin and β-catenin protein in 40 cases of hepatocellular carcinoma and 10 cases of adjacent normal tissue expression, and to analyze the expression of these two proteins HCC clinical pathological factors and the correlation between , so as to explore their role in the development of HCC. Materials and Methods 1. Collect the First Affiliated Hospital of Zhengzhou University, 1996-2006 due to liver cancer after surgical resection pathology archived paraffin blocks. Selected 40 cases of HCC and 10 cases of adjacent normal liver tissue. Where well-differentiated HCC 18 cases, moderately differentiated HCC 12 cases, poorly differentiated HCC 10 cases; lymph node metastases in 24 patients, 16 patients without lymph node metastasis. All tissue specimens were fixed in 10% neutral formalin, embedded in paraffin after routine dehydration. 2 immunohistochemistry (SP method) were detected Livin and β-catenin in HCC and adjacent normal tissue expression. 3 All the experimental data using SPSS10.0 statistical analysis software for processing, two samples was used to compare rates between the two groups in the rate of X ~ 2 test, correlation analysis using Spearman rank correlation analysis. Results 1.Livin protein expression in HCC Livin protein positive staining that is brown granules located in the cytoplasm of hepatocytes and HCC, 40 cases of Livin expression in HCC site is located in the cytoplasm, 40 cases of HCC, 31 cases, positive expression rate was 77.50%, 10 cases of normal tissues Livin weakly positive in two cases, the positive rate of 20%, compared to the two groups was statistically significant positive expression (P <0.05); Livin in well-differentiated, moderately differentiated, low- differentiated HCC, the positive expression rate was 72.22% (12/18) 75.00% (9/12), 100% (10/10), compared to well-differentiated and poorly differentiated significantly (P <0.05), with well-differentiated, moderately differentiated and poorly differentiated compared to no significant difference (P> 0.05); Livin lymph node metastasis in hepatocellular carcinoma positive expression rate was 75.00% (12/16), Livin liver metastasis in lymph nodes are not cell carcinoma positive expression rate was 79.00% (19/24), the difference between the two groups was not statistically significant (P> 0.05). 2.β-Catenin protein expression in HCC β-catenin protein was stained brown granules, located in the normal liver cell membrane, HCC tissues expressed mainly in the cytoplasm. 40 cases of HCC, 29 cases, the positive expression rate was 72.50%; 10 cases of normal tissues in three cases weakly positive, the positive rate of 30.00%, the second group, the difference was statistically significant (P <0.05); β -catenin in well-differentiated, moderately differentiated, poorly differentiated HCC, the positive expression rate was 61.11% (11/18) 83.33% (10/12), 80.00% (8/10), there was no difference among the three groups compared statistically significant (p> 0.05); β-catenin in lymph node positive expression rate of HCC group was 50.00% (8/16), β-catenin in HCC patients without lymph node metastasis positive expression rate was 87.50% (21/24) the two groups was statistically significant difference compared (p <0.05). 3.Livin protein and β-catenin protein expression in 40 cases of HCC with β-catenin expression of Livin protein were positive and 27 cases were negative expression number is seven cases, β-catenin expression of Livin gene expression negative positive four cases, β-catenin expression negative expression of Livin in 2 cases, by spearman rank correlation analysis and inspection, both of which showed significant positive correlation (r = 0.826, P <0.05). Conclusion 1.Livin highly expressed in HCC and normal liver tissue and low expression with histological grade and lymph node metastasis, suggesting that Livin expression may be associated with HCC occurrence, development and prognosis. 2.β-catenin is highly expressed in HCC and normal liver tissue and low expression with histological grade and lymph node metastasis, suggesting that β-catenin protein is highly expressed in HCC occurrence and development. 3.Livin with β-catenin protein expression showed significant positive correlation, suggesting that they may play in the occurrence of HCC coordination role. 4.Livin and β-catenin protein expression in HCC tissue differentiation and lymph node metastasis has yet to expand the relationship between sample size, further study.
