The purpose of looking for a long-term stable Tubing sinus model of production methods and to detect the transfection of the the HCN2 gene mesenchymal stem cells for Tubing the sinus model of the . The method of Japanese white rabbits were randomized with radiofrequency ablation and formaldehyde wet dressing method produced Yat sick sinus model . By the the sternum right edge of the open-chest rabbits were anesthetized , the bulk of the electrode in the superior vena cava and right atrium junction sinus node area discharge, a decrease of 30-50% in sinus rhythm or junctional escape rhythm for successful ablation flag and formaldehyde wet packing method as controls. Rabbit after the two methods was observed changes in heart rate and intrinsic heart rate were measured . Mesenchymal stem cells cultured in vitro , the extract containing HCN2 and GFP plasmid and plasmid transfection into mesenchymal stem cells injected into sick sinus rabbit right atrium , the control group injected with saline , observed transfection HCN2 gene between mesenchymal stem cells Tubing sinus model . Results of radiofrequency ablation produced the Tubing sinus model than formaldehyde the wet compress law group, heart rate and intrinsic heart rate slowed down , formaldehyde the wet compress law group than there is no significant difference in comparison with the normal group . Injection containing the HCN2 gene mesenchymal stem cells of rabbit heart rate compared with the control group , no significant difference . Conclusion formaldehyde ablation , radiofrequency ablation production the Tubing sinus model is a more optimized . Rabbit right atrium instead of to build the appropriate biological pacemaker injection sites .
