Abstract


Anthrax caused by Bacillus anthracis is a potent zoonotic infectious diseases , extremely hazardous . Especially in recent years , international terrorists from using the anthrax bacillus spores manufacturing terrorist event , seriously interfere with people's daily lives and the social and economic order . Therefore , its study has some theoretical value and the potential social benefits . In the present study , the use of the principle of plasmid incompatibility , A16R and A16 of Bacillus anthracis , respectively as the starting strain of experiments , a pXO1 plasmid was constructed the deletion mutant A16RPI1 and the pXO2 plasmid deletion mutant A16PI2 . Growth trends , glucose metabolism on the the strains A16RPI1 A16RP and A16PI2 and A16R , and has identified A16PI2 the capsular generation capability ; were prepared 23 h A16RPI1 with A16RP whole cell protein samples and A16PI2 and A16R 20 h , whole cell protein samples , pH 4-7 gradient two-dimensional electrophoresis to identify differentially expressed proteins and protein spots were mass spectrometric detection analysis . Get the following results : 1 ) of strain the A16RPI1 with strains A16RP of the growth trend , and also no difference between the two sugar metabolism . 2 ) the strains A16PI2 strain A16R growth trend is consistent , but the sugar metabolism slightly different the , A16PI2 metabolism of fructose but the A16R they can not metabolize fructose . After the culture experiments capsulatum , A16PI2 does not generate the capsule. 3) the strains A16RPI1 with The strain A16RP stable protein at pH 4-7 gradient five differentially expressed proteins , these proteins are located in the cytoplasm and is in the process of translation elongation factor and amino acid metabolism enzyme . 4) the strains A16PI2 strain A16R stable phase protein at pH 4-7 gradient of 22 differentially expressed proteins the basic positioning these proteins in the cytoplasm , only one protein of unknown function is located in the cell membrane , their main function the process of translation , transcription regulation , transport and metabolism of amino acids and Bacillus generate relevant .
