Abstract


In order to further understand the WSSV (White spot syndrome virus WSSV) protein VP37's role in the process of virus attachment, electrophoretic elution VP37 recovery and preparation of its polyclonal antibody. The primary cultured shrimp blood lymphocytes as a matrix, observed VP37 polyclonal antibody combination of WSSV and cell inhibition. VP37 polyclonal antibody against WSSV membrane protein binding inhibition and verified in the microtiter plate. The experimental results show, a polyclonal antibody of VP37 WSSV and cell combination having certain inhibition show that VP37 WSSV adhesion protein (virus attachment protein) or one of its components. The primary cultured shrimp blood lymphocytes as a matrix of five monoclonal antibodies by enzyme-linked immunosorbent assay (Enzyme Linked Immunoserbent Assay, ELISA) test: 1A4, 6A4, 2B2, 4A1, 1D5 neutralization capacity screening identified two can be partially inhibit WSSV a monoclonal antibody binding to cells, NO were 2B2 and 1D5, 2B2 in better than 1D5, and the remaining three are not observed and effects. Complete particles of WSSV sodium dodecyl sulfate - polyacrylamide gel denaturing electrophoresis (sodium dodecyl sulphate-of polyacrylamide - Gel electrophoresis to SDS-PAGE), and its high molecular weight protein profiles can be obtained, can be observed from the upper 7 protein molecular weight: 252.1,207.2,201.7,172.8,156.9,128.8,105.7 kD, and analyzed by electrophoresis pattern analysis software GelproAnalysis on their relative content of the content in descending order of 252.1,105.7,128.8,201.7 , 207.2,172.8,156.9 Kd. Western blot using nitrocellulose membrane (Protein Blotting) and alkaline phosphatase chromogenic substrate were initially identified two of the high molecular weight range and shrimp gill membrane protein binding protein molecular weights were 157Kd and 233Kd , reveals these two proteins may be part of the virus-binding protein. After working on the VP37 protein-terminal sequencing, and thus its role in the the WSSV life processes of research from the gene level. The experimental results show that the VP37 end closed, it needs to be treatment before sequencing. Adhesion protein ultimately determine the need to go through the body and in the experiments, this need its antibody, was prepared by the in vivo neutralization test of the individual level to verify the role of adhesion proteins.
