Abstract


Objective To investigate the enteric nervous system changes in rats with severe acute intestinal motility and Cheng Qi Tang Chaishao on Intestinal nitric oxide synthase (nNOS) expression. Methods 30 SD rats were randomly divided into three groups: sham operation group, SAP model group Chaishao, Cheng Qi Tang the group (CSCQD group); SAP model group CSCQD of group by retrograde cholangiopancreatography injection mass concentration of 50 g · L- 1 sodium taurocholate rat model of SAP, respectively intragastric administration of saline and liquid CSCQD. Rats according to the upright abdominal X-ray, the determination of the gastrointestinal transport index (TI), HE staining under light microscope to observe the pathological changes of the pancreas, observed by double immunofluorescence staining plexus in the ileum, colon myenteric plexus and colonic mucosa NOS neurons in the rat back, colon nNOS mRNA expression detected by RT-PCR method. Results Compared with the sham group, SAP model group bowel significant expansion of the delivery index was significantly lower (0.38 ± 0.0179) vs (0.23 ± 0.0156), p lt; 0.05]; compared with the SAP group, CSCQD group intestinal expansion not passed index was significantly higher (0.51 ± 0.0145) vs (0.23 ± 0.0156) (p lt; 0.05)]. Compared with the sham group, SAP model group were a large number of red or dark red turbid ascites, significant expansion of the bowel, mesentery, omentum and pancreas visible fat around the saponification, the pancreas was necrotizing changes under light microscope pancreatic tissue pathology damage is significantly increased [(1.7 ± 0.7) vs (15.6 ± 1.2) (p lt; 0.05)]; compared with the SAP group, CSCQD group ascites, bowel dilatation, mesentery, omentum, the degree of saponification of fat around the pancreas reduce the light microscope, the pathological damage of pancreatic tissue was significantly reduced (15.6 ± 1.2) vs (7.0 ± 1.4) (p lt; 0.05)]. Compared with the sham group, SAP model group back to colon myenteric plexus NOS neurons volume increases to, thickening of nerve fibers, NOS nerve yuan total percentage of neurons was significantly higher (p lt; 0.05); SAP model group, CSCQD group NOS neurons small, thin nerve fibers, NOS neurons in the total percentage of neurons was significantly lower (p lt; 0.05), three groups of rats colonic submucous plexus NOS neurons accounted for percentage of total neurons was no significant difference (p GT; 0.05) Compared with the sham group, SAP model group back colon nNOS mRNA expression increased (p lt; 0.05), compared with the SAP group, CSCQD group back to the colon nNOS mRNA expression decreased (p lt; 0.05). Conclusion SAP rat intestinal motility occurrence of enteric nervous system (ENS) remodeling, back-related the colon NOS neurons in the expression increased. With a pass in the capture, clearing and detoxifying effect CSCQD reversal ENS NOS nerve plasticity, so back to colon nNOS mRNA decreased expression of NO decreased production of intestinal smooth muscle contraction, and promote the role of intestinal motility, and ultimately reduce the SAP rat pancreatic tissue injury
