Objective: burdock (Arctium lappa L.) is a the Asteraceae Burdock species. Fruit Arctium of legal drugs, its roots as a food to be welcome in Japan, has a potential role in cancer prevention. 60s of the last century, Japanese scholars have burdock root extract polyacetylenes, recent studies show the polyacetylenes are likely to have the biological activity of the anti-tumor. The purpose of this pilot study is to explore whether active ingredient in the prevention and treatment of tumors, to lay the foundation for further research and development of burdock root burdock root. Method: ① burdock root pharmacognosy identification using conventional sensory method and microscopic identification method. The identification of the chemical composition using thin layer chromatography, chloroform - methanol - water (40:8:1) as developing agent, set UV light (254nm), under review. Test sample preparation using liquid - liquid partition extraction. Burdock root slices, room temperature, dried, extracted with 95% ethanol, solvent recovery. Alcohol extract the aqueous suspension, washed with petroleum ether, chloroform, ethyl acetate, n-butanol extract, together with the post-extraction aqueous layer (and suspended matter) separately recovering the solvent to give the corresponding 50-part sample of the different polarities. (2) sample anti-tumor activity were evaluated using the MTT assay in vitro tumor cell growth inhibition test (the mouse tumor HepA and S180), evaluation of the sample IC50; (3) samples immune activity evaluation vitro Con A-induced lymphocyte proliferation test, evaluation index for the ⑤ samples chemotherapy on lymphocyte proliferation (or inhibit) rate; ④ samples attenuated activity was evaluated in vitro tumor chemotherapy drug doxorubicin, cisplatin destruction of bone marrow cells in vitro test model, the observed protective effect of samples of of killing bone marrow cells; evaluation of synergies to the proliferation of tumor cells in vitro inhibition test to observe the impact of the combined sample of the chemotherapy drug doxorubicin IC50; evaluation of the the ⑥ anti-cancer effect of using the Ames test, test strains TA 98, select the chemotherapy drug doxorubicin factors as positive mutagenic agent to adriamycin to produce mutagenic effects and on this basis, to observe the sample the presence or absence of anti-mutagenic activity. Results: ① burdock root fresh goods as the cylindrical fleshy root, skin dark brown, wrinkles. The fleshy gray, slightly bitter taste sticky sex. The moisture content of the fresh products of approximately 83.60%. Detailed features under the microscope for cork layer several rows of cells wide cortex, phloem elongated triangle, broad bast ray; the cambium significantly; xylem vessels more radially arranged unmyelinated; common near the center of fracture. Chemical analysis showed that the unique the anticancer active ingredients Arctiin burdock root without Arctium. Petroleum ether, chloroform, ethyl acetate, n-butanol and the aqueous layer extract yield was 0.20%, 0.18%, 0.40%, 4.08%, 13.55%, respectively. The ② burdock root extract various parts mice Hep A, S180 tumor cells have different levels of cytotoxicity, in a certain concentration range remained essentially clear dose-effect relationship. Burdock root petroleum ether, chloroform, ethyl acetate, n-butanol, water extract of in hepatoma Hep A cells of IC50 for 25.97,30.62,40.67,81.80,988.81 μg / mL, respectively. IC50 on S180 cells were 9.42,13.30,50.71,32.18,2395.80 μg / mL. Petroleum ether, chloroform, extracts shows stronger cytotoxicity, the cytotoxic effect of the water extract is very low. ③ burdock root extract various parts of the Con A-induced mouse spleen lymphocytes (T lymphocytes), value-added showed a clear inhibitory effect, its basically a dose-dependent inhibition of lymphocyte proliferation. Burdock root petroleum ether, chloroform, ethyl acetate, n-butanol, water extract of lymphocyte proliferation inhibition IC50 for 26.67,28.68,27.26,156.88,629.23 μg / mL, respectively. ④ burdock root extract and water extract of mouse bone marrow cells can be induced by adriamycin injury has a protective effect of n-butanol, n-butanol extract slightly better than the water extract, n-butanol extract concentration of 10,40 μg / mL, clear against doxorubicin killing effect on bone marrow cells improve the survival rate of cells of the bone marrow cell survival increased from 41.71% to 45.07%, and 49.40% (P lt; 0.05). Sample concentration 2.5μg/mL can clearly reduce cisplatin induced bone marrow cytotoxicity. The cell survival rate of 61.78% to 66.59% (P lt; 0.05). Water extracts of burdock root weaker protective effect of bone marrow cell damage induced by cisplatin showed a protective effect only in the low-dose (2.5μg/mL). ⑤ burdock root petroleum ether extract (3μg/mL), chloroform extract (3μg/mL) in combination with doxorubicin, doxorubicin on tumor cells of mice Hep A IC50 were 3.76,3.11 μg / mL with single use the doxorubicin IC50 (4.69μg/mL) compared decreased, but not statistically significant. The ethyl acetate extract (5μg/mL) in combination with doxorubicin, doxorubicin mouse Hep A tumor cytotoxic IC50 increased to 5.39μg/mL, no statistical significance. ⑥ burdock root n-butanol extract and water extract of the test concentration 0.1,1.0,10.0 μg / dish, can reduce doxorubicin mutagenic effect of the TA 98 strain, mutagenic inhibition rates were 31.15 65.41,69.78% and 34.43,60.93,67.25%, showed a dose-effect relationship. N-butanol in the high dose group of the extract and water extract in the high-dose group compared with the doxorubicin group, the difference was statistically significant (P lt; 0.05) shows that doxorubicin induced becomes moderate inhibition. Conclusion: The chemical composition of burdock root and Arctium differences, the former does not contain known to have anti-tumor activity of chemical substances Arctiin. The burdock root having anti-tumor active ingredient, having able to resist the bone marrow cells of the chemotherapeutic drugs doxorubicin and cisplatin cytotoxicity in component addition burdock root also contain ingredients having anti-mutagenic effect. Burdock root with the value of research and development as a cancer prevention drug or food.
