Objective: Dunaliella (Dunaliella salina) due to its high carotene content, anti-aging, anti-cancer, reduce the incidence of cardiovascular disease, skin care and vitamin A supplementation, β-carotene contained in Dunaliella Dunaliella research and development is now hot, Dunaliella other compounds (polysaccharide) of biological activity at home and abroad yet reported. The purpose of the test is to explore Dunaliella whether in addition to the β-carotene and polysaccharide also contain other active ingredients to cancer prevention and control, to be more fully developed the use of lay the foundation for Dunaliella. Methods: ① Dunaliella extract preparation: Dunaliella paste washed with distilled water to remove salt, dark conditions, dried and developed into a powder. The dried alga powder was washed with petroleum ether → chloroform → ethyl acetate → acetone → ethanol → Water reflux extraction, according to the polarity obtained by ascending the six parts of the extract, the water extracts by ethanol precipitation (water / 1:5, V / V) aqueous ethanol, to give two samples; chloroform extract was purified by silica gel column chromatography after removal of the β-carotene to give three samples received a total of A, B2, B3, B4, and C , D, E, F1, F2 nine for the experiment with the sample. Rating: ② sample anti-tumor activity was evaluated by MTT assay samples of the tumor cells (mouse tumor U14 and S180 human tumor cell A549 and Hela) cytotoxic activity, evaluation samples IC50; ③ samples immune activity evaluation: in vitro ConA induced lymphocyte proliferation test, evaluation of chemotherapy drugs promote (or inhibit) rate; ④ samples Attenuation of evaluation: a cancer chemotherapy drug epirubicin destruction of bone marrow cells in vitro test model, the sample was observed on lymphocyte proliferation the protective effect of the destruction of bone marrow cells, the evaluation of bone marrow cell survival; the ⑤ samples chemotherapy synergy evaluation: the proliferation of tumor cells in vitro inhibition test, the sample was observed with the chemotherapy drug epirubicin, cisplatin combination of tumor cell inhibition rate impact evaluation for tumor inhibition rate and the Q value; evaluation of the the ⑥ anti-cancer effect: the Ames test, select TA98 test strain, chemotherapy drug doxorubicin positive mutagen doxorubicin mutagenic effects and samples with or without anti-mutagenic effects observed on this basis, the MR value evaluation. Results: ① 600g Dunaliella paste washed to remove salt to get 131.1g Duchenne algae dry powder. The extract objects A, B (β-carotene and B2, B3, B4), C, D, E and F (F1 and F2) yield of 5.19%, 10.53%, 0.92%, 0.38%, 4.27%, 11.67%. UV spectrophotometric spectrophotometry and thin-layer chromatography analysis to extract B containing β-carotene, β-carotene 146.2mg by column chromatography. ② Dunaliella partial extract of mouse U14 the S180 and people A549, Hela tumor cells beads have different levels of cytotoxicity remained essentially clear dose-effect relationship in a certain concentration range. Wherein A, B3 showing strong cytotoxic activity, Fl, F2, without cytotoxic activity (IC50 GT; 1.0mg/mL). Value added ③ Dunaliella partial extract of ConA-induced mouse spleen lymphocytes (T lymphocytes) showed clear inhibitory effect, its basically a dose-dependent inhibition of lymphocyte proliferation. B4 and C ④ extracts on doxorubicin-induced injury of mouse bone marrow cells have a protective effect, in which the role better than B4 C the extract C final concentration and for 2.5,5,10,20,40 μg / mL of when against the killing effect of doxorubicin on bone marrow cells of the bone marrow cells survival were increased from 47.46% to 57.62%, 55.11%, 60.35%, 56.71% and 57.85% (P less than 0.05). ⑤ Dunaliella extract A and B3 with cisplatin combination of U14 tumor cell inhibition has a synergistic effect (Q gt; 1.15), but the two samples with Epirubicin no significant synergistic effect of the mouse U14 cells suppression ( Q = 0.93-1.05), while on S180 cells showed some antagonism (Q lt; 0.85). ⑥ Dunaliella extract F1, the test concentrations of 0.1, 1.0, 10.0 μg / (?), Can antagonize the mutagenic effect of adriamycin on the TA98 strain mutagenesis inhibition rates were 53.38,94.79,66.07%. F1's low in the high-dose group compared with doxorubicin induced group, the differences were significant sex (P lt; 0.05). Conclusion: Dunaliella in addition to β-carotene and polysaccharide also contains other anti-tumor active ingredient, can resist the chemotherapy drug epirubicin due to the destruction of the active ingredient of the bone marrow cells and improve tumor cells to chemotherapeutic drugs cisplatin sensitivity ingredients, in addition Dunaliella ingredients also contains anti-mutagenic effects. Dunaliella as having the value of the research and development of cancer prevention and treatment of drug or food.
