Abstract

This experiment experimental means of transmission electron microscopy, Camponotus japonicus as experimental materials, its organizational structure, the morphological characteristics of the antennal lobe olfactory ball observed. In addition, the conventional embedding method using epoxy Epon812 antennal lobe of immune colloidal gold experiment, and immune colloidal gold electron microscopy made a preliminary study. The experimental results and discussion are as follows: 1. Antennal lobe ultrastructural analysis: the antennal lobe is the primary olfactory center of insects, and olfactory ball is a subunit of the antennal lobe morphology and function, its organizational structure and invertebrates basically consistent. Therefore, starting from the the insect olfactory ball structure function study their brain activity, provide a theoretical basis for the study of vertebrate brain. So as to promote the study of the higher animals brain physiology, pathological mechanisms and treatment. In this experiment, Camponotus japonicus as experimental material, its organizational structure and morphological characteristics of the antennal lobe olfactory ball and its surrounding tissue in the ultrafine level. The results found that: (1) Camponotus japonicus antennal lobe distributed in many different sizes, the olfactory ball nearly spherical. Ball distribution of these fibers is not continuous, the neuron cell population, a non-olfactory fiberballs fiber separately. The periphery of the olfactory ball more or less by the number of glial cells or protrusions surrounding. (2) olfactory ball is composed of nerve fibers nerve fiber network, a large number of synaptic connections between nerve fibers. In addition, these fibers were also observed within the mitochondria, endoplasmic reticulum organelles. A complex antennal lobe brain structure reflect the Camponotus japonicus developed olfactory system. 2. Immune electron microscopy (SEM): immune colloidal gold electron microscopy experiments can understand the the insect brain neurotransmitter qualitative, quantitative distribution, which is the basis for further explore the neural activity patterns. We worked out through access to the extensive literature application embedded in epoxy resin combined immune colloidal gold electron microscopy experimental route. The biggest advantage of this method is that after polymerization embedding block is easy to cut into thin slices, almost no chatter marks generated. The standard gold results show that some synaptic activity area was the positive reaction of γ-aminobutyric acid (GABA), the electron microscope, centralized distribution of colloidal gold particles. Some synapses in the presence of a large number of synaptic vesicles, but only a small portion of vesicles was the GABA positive reaction, indicating where GABA is the total of neurotransmitters (co-transmitter), there are other neurological material existence, the regulation of nerve activity.
