Ray 's Macrothele (Macrothele raveni) is our recent identification of a new species of spider , launched a study of the venom of the spider . Isolated a novel polypeptide neurotoxin Raventoxin-Ⅱ, completion of a structural analysis, carried out a preliminary toxicity study of Raventoxin-Ⅱ , the results are as follows : 1 . First to isoelectric focusing using immobilized pH gradient strips (pH3-10) , the choice of 12.5% ??separating gel second SDS electrophoresis , 2D-PAGE electrophoresis pattern obtained raveni spider venom ; using MALDI -TOF mass spectrometer , mass spectra raveni spider venom ; they show the distribution of the spider venom peptide and protein components of the type and number as well as molecular weight and isoelectric point range . 2. By cation exchange chromatography , RP-HPLC purification of a novel peptide toxins from the venom of mice to produce mice by intraperitoneal injection , difficulty in breathing , paralysis and death . 3. By anion exchange chromatography , RP-HPLC purified a novel peptide toxins from the venom , named to as Raventoxin - II . Raventoxin - II mice can death, showed weak mammal toxicity . By mass spectrometry , its precise molecular weight of 3021.56Da. The amino acid composition analysis and amino acid sequence analysis showed that a Raventoxin - II consists of 33 amino acid residues consisting of, containing 6 cystine , formation of three disulfide bonds . Its sequence is as follows : H 2 NICADEGGPCVAGIGCCAGLR-CSGAILGLAGSCQ. Homology with the rest of the currently known spider toxins found Raventoxin-Ⅱ minimal protein database search .
