Abstract


Free hair transplantation of donor origin on the clinical problem of insufficient human hair hair follicle microdissection , culture , focusing on observation of morphological changes of cultured hair follicle in the late Changes to investigate the in vitro growth of hair follicles and periodic regeneration by summarizing the experimental operation in the hair follicles separation technology and culture conditions suitable for the separation of the hair transplant hair follicles , save method and influencing factors provide a reference for further allogeneic hair transplant and hair tissue engineering research ; the improvements provide clues in order to obtain better results . Methods: under aseptic conditions , take beauty rhytidectomy resected scalp Microdissection free single hair follicle and cultured in DMEM medium containing 10% fetal calf serum , continuity dynamic observation of hair follicle growth rate and morphology changes of more than 30 days , and histological observation taken the hair follicle paraffin sections before and after culture , contrast , record results and photographed . Results: We have successfully carried out four batches of human hair free hair follicles cultured in vitro ; , hair follicles in cultured containing calf serum DMEN , the slower growth rate , early entry into the degradation period ; 3 cultured hair follicles into the degradation period after dermal papilla cells (DPC) there are two different distribution : ① most of the hair follicles the DPC crunch into a group located in the middle of degraded hair follicles lower edge ; ② and about 30% of the hair follicles the DPC out free adherent growth epithelioid cells and dermal sheath cells around to radiating from growth ; 4 , free hair follicle paraffin sections improved method of making the same good results . Conclusion : 1 . Human hair free hair follicles in vitro culture , media constitute a direct impact on the growth of hair follicles . The growth of hair follicles in the DMEM medium containing fetal calf serum is easy to enter the degenerative phase ; 2. Cultured human hair follicles in culture late into the degradation period , there are two distribution types ; dermal papilla cells . Compared with the previous method , we improved the hair follicle producer method simplifies J ' steps to obtain the the same effect.
