Objective: 1. Research on the use of serum pharmacology of anti-tumor drugs tumor sanjie Dan on human cervical carcinoma cell line HeLa cell growth inhibition and induction of apoptosis and cell cycle. 2. Investigate the tumor sanjie Dan possible mechanism of antitumor effect. 3. Dan's understanding of the tumor sanjie acute toxicological effects. Methods: 1. In accordance with the requirements of serum pharmacology preparation containing serum: Dan will be made of the tumor sanjie water decoction orally to rabbits, acquire and rabbit blood serum was separated. 2. Cell culture: The human cervical carcinoma cell line HeLa cells were inoculated with 10% fetal bovine serum in RPMI-1640 medium at 37 ℃ with 5% CO_2 the incubator. 3. Choose serum containing different concentrations of human cervical cancer cell line HeLa cells together 24 hours, using an inverted microscope morphological changes; using MTT reduction assay optical density (OD value), inhibition rate was calculated; using flow cytometry (FCM) detection of tumor-induced apoptosis in HeLa cells Dan sanjie effects as well as effects on the cell cycle. 4. Acute toxicity test in mice by oral gavage administration mode, continuous observation of seven days, the number of animal deaths statistics, access to relevant experimental data and with the modified Karber method for the determination of their LD 50 . Results: 1. HeLa cells were treated with drug-containing rabbit serum for 24 hours, the growth of adherent gradually turned off. Many cell culture medium resuspended, the cells became round, the volume reduced, significant gaps between cells. While the control group and the saline group adherent cells grew well, the adjacent cells fused, showing polygonal, medium no or only see a little cell debris. 2. Serum containing large, medium and small dose group inhibition rates were 56.10%, 48.37%, 34.31%, significantly higher than the control group and the saline group (P <0.05); and the high-dose group was significantly higher than that in the low dose group ( P <0.05). 3. Serum containing large, medium-dose group induced apoptosis percentages were 21.58%, 13.32%, and compared to the control group and the saline group were significantly different (P <0.05); Furthermore, compared to the control group and the saline group, Chinese large, medium-dose group were so G 0 / G 1 phase cells decreased significantly, G 2 / M phase cells increased significantly, S phase cells significantly reduced by different degrees of cell cycle arrest at G 2 / M phase; small dose group makes G 0 / G 1 phase cells significantly increased, G 2 / M phase cells was significantly reduced, S phase cells decreased significantly. 4. The tumor sanjie Dan oral gavage LD 50 is: 10.11 g / kg; 95% confidence limit: 9.41 ~ 10.81g/kg conclusions: 1. The tumor sanjie Dan (containing serum) inhibits human cervical carcinoma cell line HeLa cell growth; 2. The tumor sanjie Dan (containing serum) large, medium-dose group can induce apoptosis in HeLa cells; 3.
