Objective To explore the effect of melatonin(MT) and dexamethasone(DXM) on the expression of nuclear factor- k B(NF- k B) in bronchial asthmatic rats model.Methods 32 Sprague-Dawley(SD) rats were randomly divided into four groups of 8 each: (1)Asthmatic group(Group A): Rats were injected intraperitoneally with 1ml of 10% ovalbumin(OVA) and 100mg of solution of A1(OH)3 .After 14 days,the rats were inhaled with an aerosol of 1% OVA for 20 minutes for 7 days every day; (2) DXM prvention group(Group B): As above, just before the rats were inhaled with an aerosol, 40mg/kg DXM were injected intraperitoneally; (3) MT prvention group(Group C): As Group A, just before the rats were inhaled with an aerosol, 10mg/kg MT were injected intraperitoneally; (4) Control group(Group D): As Group A, just with NS stand for OVA. 6 hours after last challenge, airway responsiveness, total cell counts and cell differential percentages were examined and levels of nitrate in bronchoalveolar lavage fluid(BALF) were detected. At the same time, inducible nitric oxide synthase(iNOS) and constitute nitric oxide(cNOS) activity levels in the lung tissues were examined, and the changes of the expression of NF-kB in the lung tissues were observed by immunohistochemical staining.Rsults 1. After OVA challenge, Group A had increased significantly total cells in BALF compared with the other groups(p<0.05); Group B and Group C had decreased significantly total cells in BALF compared with Group A, but Group C had increased significantly total cells in BALF compared with Group D(p<0.05); 2. Group A hadincreased significantly airway reaction compared with the other groups(p<0.01); Group B and Group C had decreased significantly airway reaction compared with Group A(p<0.01); 3. Group B and Group C had decreased significantly amounts of NO2-/ NO3-in the BALF and of iNOS levels in the lung tissues compared with Group A(p<0.01), but Group C had inceased significantly compared with Group D; It was found that there was no marked alteration of cNOS activity in four groups. Elevation of iNOS in the lung tissues was correlated with NO2-/ NO3- in the BALF(p<0.01); 4. The expression of NF- K B in lung tissues in Group A had increasd significantly compared with the other groups(p<0.01), but Group C had increasd significantly than Group D.Condutions There is increased the expression of NF- K B in lung tissues of asthmatic rats, the increasd activity of iNOS by NF- K B regulated could cause increased prouduction of NO, and probably cause cytotoxicity and mediate airway hyperresponsiveness. DXM and MT could inhibit activation of NF- KB in the allergic model, possibly through which the anti-inflammatory action was exerted.
