Abstract


Extraction of glutamic acid in the mother liquor of this thesis isoelectric . Bacterial cells in the mother liquor of glutamic acid isoelectric first removal . The results show that the use of sodium polyacrylate as flocculant , added 2% , temperature 60 ° C , a pH of 3.6 , stirring speed 20r/min stirring 15min bacterial removal rate of more than 90% . Second glutamate bacterial proteins as raw material , compound amino acid hydrolyzate after dilute acid pretreatment , hydrolysis with protease . By orthogonal experiment to determine the enzymatic optimum conditions : the enzyme dosage 400U / g, temperature of 40 ° C , pH 7.5 , hydrolysis time 12h, substrate concentration ratio of 1:30 . Under this condition , the measured hydrolysis rate was 72.4% of the bacterial proteins . Again , to determine the best process of desalination , and glutamate secondary electric crystalline explore . The experiments show that the glutamic secondary isoelectric crystallized in a yield of more than 70% . Additionally, this article was measured in the four kinds of systems , i.e. pure water , the solubility and the solubility curve of the bacterial solution of 1 % of NH4 concentration solution , 0.1% , and practical industrial fermentation liquid system glutamate . Fitting these curves with mathematical expressions , glutamate isoelectric point calculate the metastable zone width . Experimental results show that the width of the metastable zone in pure water , a 0.1% bacterial solution , the solution and the practical industrial fermentation broth of 1 % of NH4 concentration gradually narrowed , i.e. glutamate In these systems , the solubility of gradually becomes larger , and its fitting saturation and super saturation curve has the following form : Glutamate solution was measured at different cooling rate , stirring speed crystallization metastable zone width and different saturation of the crystal growth rate and crystal nucleation induction period . Experimental results show that the glutamic acid crystals were metastable zone is widened, with the increase of the cooling rate , stirring speed increases narrowed . The crystal growth rate increases with the relative increase in saturation increase , and the crystalline nucleation induction period with the relative increase in the degree of supersaturation is shortened .
