Abstract


ObjectiveIn order to investgate tumor growth inhibiting effect of bearing Lewis cancer mice by indomethacin (IN), one of non-steroid anti-inflammatory drug (NSAID), and sensitization the growth inhibiting effect of radiotherapy and chemotherapeutics. Provid evidence of generally application IN cure lung cancer in clinical.MethodsOne Lewis pulmonary tumor model rat, 60 kunming rats (6-Sweeks, weighed 20±2g, male, ordinary). TNF-αradioimmunoassay kit.60 kunming rats were made into Lewis pulmonary tumor model rats. These rats were taken out randomly when the longest diameter of their tumor was about 1cm, and were divided ihto 6groups, different groups of tumor-bearing mice receive therapy of differents therapeutics:IN、IN+radiotherapeutics、IN+chemotherapeutics、radiotherapeutics、chemotherapeutics and blank. The long and short diameter were measured by sliding caliper to calculate tumor volums every day.The anti-tumor rate were calculated through tumor volumetric, tumor necrosis factor (TNF-α) contents were detected by radioimmunoassay. Treatment complete take tumor tissue to made pathology paraffin section. Therapeutic effect evaluate by gross tumor volume growth inhibiting effect、TNF and pathology paraffin section after therapy complete. ResultThe gross tumor volume of all therapeutics groups growth slowly, 5 days after treatment started. To contrast all therapeutics groups with control group, tumor growth inhibited obviously(P<0.05), the inhibition rate of IN is 37.86%, the inhibition rate of chemotherapeutics is 51.24%, the inhibition rate of radiotherapeutics is 63.29%, the inhibition rate of IN+chemotherapeutics is 71.13%, the inhibition rate of IN+radiotherapeutics is 76.86%, The tumor growth inhibiting effect of IN+radiotherapy and IN+chemotherapeutics better than others therapeutics; radioimmunoassay (RIA) measure tumor necrosis factor(TNF-α) contents of serum, before treatment all groups’contents of TNF-αwere almost same, when experiment is over, they have significant difference, To contrast all therapeutics groups with control group, the contents of TNF-αsignificant difference(P<0.05). The contents of TNF-α,IN+radiotherapy and IN+chemotherapeutics higher than chemotherapeutics and radiotherapeutics; naked eye pathology observation can see different degree necrosis in all tumor organizer center. pathology paraffin section display tumor cells cellular necrosis, the two combination therapeutics, IN+radiotherapy and IN+chemotherapeutics tumor cells cellular necrosis especially manifest.ConclusionsIN could inhibit the tumor growth of rat’s pulmonary tumor model, to promote tumor cell necrosis,to explain the therapeutic efficacy of IN to lung cancer. IN association with chemotherapeutics or radiotherapeutics application to rat’s pulmonary tumor model, therapeutic efficacy is manliest. In the experiment IN made therapeutic efficacy of chemotherapeutics or radiotherapeutics manifest,as the sensitizer effect;sensitizer is one of therapeutic measures as chemotherapeutics or radiotherapeutics, make tumor cells more sensitive to chemotherapeutics or radiotherapeutics, therapeutic efficacy will more effectively. This Conclusions provid considerable proof for clinical combined therapy of lung cancer, the result may generally application toclinical lung cancer, if it can be verification by clinical experiments.
