Abstract

Objective: To simultaneously extracted from the chickpea isoflavones and saponins were isolated and purified and analyzed and identified, to verify diabetes (diabetes mellitus, DM) mice lowering blood glucose, and lipids regulating on this basis, and observe oxidative - the impact of antioxidant status in order to explore its hypoglycemic, lipid-lowering mechanism. Methods: (1) based on the physicochemical properties of isoflavones and saponins, select the appropriate extraction solvent and extraction methods, the design of a single factor experiment and orthogonal experiment worked out simultaneously extract the optimum of CI and CS, by comparing the experimental and select The crude extract skim, the deproteinized preliminary sundries. (2) select the appropriate adsorption column based on the physical and chemical properties of isoflavones and saponins, their initial separation and enrichment of organic solvent precipitation and column chromatography, on this basis, compared to filter out the best method of purifying CI and CS products obtained satisfactory purity and absorption from the general nature of the color reaction and UV characterized in several aspects of its analysis and identification. (3) the establishment of the ideal model of DM2, to verify the CI and CS hypoglycemic, adjusting the role of lipids, and further observation of the the DM mice oxidation - antioxidant status, on this basis, to explore its hypoglycemic lipid the mechanism. Results: (1) extraction with ethanol as solvent under reflux can be effectively carried out CI and CS synchronization extraction. (2) simultaneously extracted CI and CS process as follows: 70% ethanol, solid-liquid ratio: 1:10, water bath temperature of 70 ° C, extraction time 3h, extracted twice. (3) the refrigerated centrifugation method, and the isoelectric point method is crude extracts were degreased, the deproteinized ideal method. (4) H103 resin column and HPD-600 resin column can effectively remove water-soluble impurities such as sugar, salt, enriched CI and CS, and to reach a certain purification effect. (5) polyamide column chromatography better than the chemical solvent precipitation method can better separation and purification of CI and CS satisfactory purity CI and CS products. (6) first fed a high-sugar high-fat diet, and then following the method of low doses of STZ successfully established the ideal DM2 model, which is the study of the ideal model of DM2 and its vascular complications. (7) CI and CS DM mice have a clear lowering blood glucose and lipids regulating function, the DM mice \And the role of the CS in raising serum HDL-C is particularly significant, especially the role of the CI in lowering serum TG. (8) CI and CS can enhance DM Mice with activities of antioxidant enzymes, to reduce the DM mice peroxidation content, which may be one of the mechanisms of its role in lowering blood sugar, lipids regulating. Conclusion: isoflavones and saponins may be the main ingredient for lowering blood sugar, lipids regulating chickpeas, and enhance the body's antioxidant capacity may be the main mechanism of hypoglycemic, lipid-lowering.
